DNA diagnostics of Salmonella enterica subs. enterica (focusing on serovars Enteritidis and Typhimurium) in the intestinal content of domestic pigeon (Columba livia domestica)

Abstract:

Volně žijící holubi domácí patří mezi invazivní druhy, které obývají městské prostředí společně s lidmi. Tvoří všudypřítomný aspekt městského života, nicméně mnozí si stále neuvědomují potenciální hrozbu, kterou tato zvířata pro lidské zdraví předsta-vují. Bylo zjištěno, že holubi jsou přenašeči různých patogenů, které mohou nakazit člověka například prostřednictvím výměšků svého trávicího traktu. Infikovaný holu-bí trus a moč mohou kontaminovat zdroje potravy pro hospodářská zvířata, vodní nádrže, zeleninu, ovoce, a dokonce i prach. Jedním z patogenů přenášených holuby je Salmonella enterica. Tato gramnegativní tyčkovitá bakterie je původcem sal-monelózy, onemocnění charakterizovaného silným průjmem, nevolností, horečkou, intenzivními bolestmi a v nejtěžších případech může skončit i smrtí. Séroty-py enterica a typhimurium patří k celosvětově nejrozšířenějším. Cílem této studie bylo stanovit metodu izolace DNA a analyzovat prevalen-ci Salmonella enterica subs. enterica ser. enteritidis a Salmonella enterica subs. enterica ser. typhimurium ve 120 vzorcích holubího trusu, odebraných v městské části Brno–sever, pomocí metody Real-Time PCR. Výsledky prokázaly nízkou prevalenci 0,83 %, kdy pouze jeden vzorek ze 120 byl pozitivní.

Abstract:

Wild varieties of domestic pigeons are invasive species, that cohabit with humans in urban environments. They constitute an omnipresent aspect of urban life, yet many remain unaware of the potential threat these animals pose to human health. It has been established that pigeons serve as carriers of various pathogens, which can be transmitted to humans through, for instance, the secretions of their digestive tract. Infected pigeon droppings and urine have the potential to contaminate food sources for farm animals, water reservoirs, vegetables, fruit, and even dust. One of the pathogens carried by pigeons is Salmonella enterica. This gram-negative rod-shaped bacterium is the causative agent of salmonellosis, a disease characterized by severe diarrhoea, nausea, fever, and intense pain, and in the most severe cases, it can even result in death. Serovars enterica and typhimurium are ranked among the most abundant worldwide. This study aimed to establish a method for DNA isolation and to analyse the prevalence of Salmonella enterica subs. enterica ser. enteritidis and typhimurium in 120 samples of pigeon faeces, collected in Brno-North, using Real-Time PCR. The results indicated a low prevalence of 0.83%, with only one sample out of 120 testing positive.
