Kinetic mechanism of coelenterazine-dependent luciferase reaction

Abstract:

Náplní diplomové práce je teoretické seznámení s enzymovou evolucí a promiskuitou, ale také podrobný popis enzymové kinetiky včetně pokročilých technik tranzitní kinetiky, jež jsou k detailní analýze mechanismu enzymové reakce využívány. V návaznosti na teoretický popis byla dále provedena detailní kinetická charakterizace enzymu luciferasy z organismu Renilla reniformis pomocí metod zastaveného toku a rychlého zhášení. Výsledkem byl prvotní návrh kinetického mechanismu koelenterazin-dependentní luciferasové reakce. Získané poznatky jsou přitom prvním krokem k detailnímu popisu reakčního mechanismu, jehož vyřešení může přispět k lepšímu pochopení divergentní evoluce nových enzymatických funkcí, ale může mít i velký dopad na všechny výzkumné a průmyslové oblasti, kde se luminiscenční eseje využívají.

Abstract:

This diploma thesis provides the basis of enzyme evolution and promiscuity, and a deeper discussion of enzyme kinetics including the advanced techniques of transient kinetics available for a detailed analysis of enzymatic reaction mechanisms. In connection with the theoretical background, a detailed kinetic characterisation of the luciferase from Renilla reniformis was performed using the methods of stopped-flow and rapid quench-flow. As a result, the very first minimal kinetic mechanism of the coelenterazine-dependent luciferase reaction was proposed. The results are very important for a detailed understanding of the reaction mechanism, which can have a high impact on the many fields in which the luminescence assays are applied. Moreover, completing the missing information about the mechanism of the luciferases can lead to a valuable understanding of divergent evolution of new enzymatic functions.
