The effect of auxin herbicides on the regulation of endogenous auxin homeostasis in plants

Abstract:

The effect of auxin herbicides on endogenous auxin homeostasis have not been studied detailly yet. The aim of this master thesis is to reveal the auxin background of model plant Arabidopsis thaliana after application of five herbicides in different concentrations. The experiments were made on seedlings growing on medium with chosen herbicides and on mature plants which were sprayed with the herbicides. The auxin profiles were established using liquid chromatography combined with tandem mass spectrometry (LC-MS/MS). According to changing auxin profiles the target genes were selected and tested using real time - quantitative PCR (RT-qPCR). After auxin herbicide treatment the changes in gene expression were observed and changes in auxin profiles were observed as well, especially decreasing of endogenous IAA levels, changes in IAA precursor levels and increase of auxin conjugates and degradation products.

Abstract:

Účinkem auxinových herbicidů na endogenní auxinovou rovnováhu rostlin se doposud detailněji nikdo nezabýval. Tato diplomová práce si proto klade za cíl rozkrýt auxinové pozadí modelové rostliny A. thaliana po aplikaci pěti auxinových herbicidů v různých koncentracích. Experimenty byly prováděny na klíčních rostlinách rostoucích na médiu obsahující příslušný herbicid, ale také na dospělých rostlinách, které byly herbicidy sprejovány. V testovaných rostlinách se pak pomocí kapalinové chromatografie sdružené s tandemovou hmotnostní spektrometrií (LC-MS/MS) podařilo stanovit celkové auxinové metabolické profily. V návaznosti na zaznamenané změny auxinových profilů byly vytipovány kandidátní geny, které byly testování pomocí RT-qPCR metody. Po ošetření auxinovými herbicidy byly pozorovány jak změny na urovni genové exprese tak i změny auxinových profilů, zejména pak snižování endogenní hladiny IAA, změny hladin prekurzorů biosyntézy IAA a nárůst produktů konjugace a degradace IAA
