The Treatment of Breeding Material from Tomato Brown Rugose Fruit Virus by In Vitro Methods

Abstract:

Diplomová práce zpracovává problematiku karanténního škodlivého organismu viru Tomato brown rugose fruit virus (ToBRFV) na porostech rajčat a paprik. Výsledkem práce mělo být ozdravení papriky seté (C. annuum L.) v podmínkách in vitro s využitím metod termoterapie, chemoterapie a meristémových kultur. Metoda termoterapie byla na papriky aplikována při teplotě 35 °C po dobu tří týdnů. Pro metodu chemoterapie byly použity antivirotické látky (5-azacytidin, 2-thiouracil a ribavirin) v osmi variantách koncentrací. Dále byl proveden převod vrcholových apikálů po metodě chemoterapeutické, který potenciálně zakládal nové explantáty bez přítomnosti viru. Detekce metodou RT-qPCR po technikách ozdravování prokázaly přítomnost viru ToBRFV ve všech testovaných vzorcích. Výsledky práce potvrdily vysokou infekčnost viru ToBRFV. Veškeré postupy práce během ozdravování byly realizovány v rámci Izolačního zařízení Mendelea – ústavu genetiky (Zahradnikcá fakulta, MENDELU), které má povolení pro uchovávání a manipulaci s tímto karanténním škodlivým organismem.

Abstract:

Diploma thesis deals with the problematics of quarantine organism of Tomato brown rugose fruit virus (ToBRFV) on tomatoes and peppers. The result of this work should be treatment of bell pepper (C. annuum L.) in in vitro conditions with the use of methods thermotherapy, chemotherapy and meristem cultures. The thermotherapy method was applied to peppers at a temperature of 35 °C for three weeks. For chemotherapy method were used antivirotical substances (5-azacytidine, 2-thiouracil and ribavirin) in eight variants of concentrations. Further it was transfered apical top after chemotherapy method which potentially established new explants without virus. Detections by RT-qPCR method after treatment techniques proved presence of ToBRFV virus in all tested samples. Results of this work confirmed high infectivity of virus ToBRFV. Entire work approaches during treatment were realized within the Isolation Facility of Mendeleum – Institute of Genetics (Horticulture faculty, MENDELU), which has a authorization for preservation and handling with this quarantine organism.
