Characterization of Hyaluronan Using Gel Permeation Chromatography

Abstract:

Diplomová práce se zabývá stanovením molekulové hmotnosti a distribuce molekulových hmotností komerčních vzorků hyaluronanu. K tomuto účelu byla použita metoda gelové permeační chromatografie (GPC). Pozornost byla věnována nalezení chromatografických podmínek vhodných pro charakterizaci hyaluronanu a posouzení vlivu složení mobilní fáze na chromatografickou separaci tohoto biopolymeru. Bylo zjištěno, že chování vzorku hyaluronanu a následně hodnoty molekulových hmotností Mw a Mn jsou závislé na typu soli obsažené v mobilní fázi i na typu standardu použitého pro kalibraci chromatografické kolony.

Abstract:

In this graduation thesis, determination of the molecular weight and molecular weight distribution of commercial samples of hyaluronan was performed. For this purpose, gel permeation chromatography (GPC) was employed. The thesis is also focused on finding the chromatographic conditions suitable for the characterization of hyaluronan by GPC and on the assessment of the influence of mobile phase composition on the chromatographic separation of this biopolymer. It was found out that the behaviour of the hyaluronan and values of Mw and Mn are dependent on the type of salts contained in the mobile phase and on the type of standard used for the calibration of the chromatographic column.
