Population genetic structure, genetic diversity and natural history of Anemone sylvestris (Ranunculaceae) inferred from nuclear microsatellite data

Abstract:

Práce se zaměřuje na ohrožený druh sasanku lesní Anemone sylvestris. Anemone sylvestris je druhem suchých trávníků a prosvětlených lesních lemů, v současnosti velmi ohrožených stanovišť. Pro účinná ochranářská opatření je nezbytná znalost genetické diverzity populací. Cílem práce je proto popsat populačně-genetickou strukturu tohoto druhu na území České republiky, včetně stanovení míry příbuznosti jednotlivých populací a jejich potenciálního genetického původu. Všechny tyto charakteristiky byly stanoveny pomocí mikrosatelitových DNA markerů.Prostřednictvím molekulárních analýz bylo ukázáno, že populace v České republice jsou ve srovnání se zahraničními populacemi velmi geneticky chudé. Zároveň jsou však navzájem výrazně odlišné, a tedy nedochází zde k žádné nebo velmi malé výměně genů mezi populacemi. Především v populacích v České republice hraje významnou roli vegetativní rozmnožování, které umožnilo fixaci převážné části lokusů v heterozygotní podobě a zamezilo tak absolutní ztrátě genetické diverzity vlivem genetického driftu. Zároveň naprostá převaha jediného klonálního genotypu vlivem kompetice v každé populaci ještě více posílila vzájemnou odlišnost i velmi geograficky blízkých populací. Téměř každá populace je tak tvořena samostatnou genetickou linií.Naprostá odlišnost populací spolu se zcela náhodnou fixací alel v populacích jsou pravděpodobně pozůstatky velké genetické diverzity původně velké populace Anemone sylvestris v minulosti. Její jednotlivé části byly fixovány právě prostřednictvím klonů.

Abstract:

This thesis focuses on an endangered species of Anemone sylvestris. Anemone sylvestris is found in dry grasslands and bright woodland edges, both of which are currently at high risk. Knowledge of genetic diversity of populations is essential for the application of effective conservation measures. The aim of this thesis is to describe the population-genetic structure of this species in the Czech Republic, including the determination of the degree of affinity of individual populations and their potential genetic origin. All of these characteristics were determined by microsatellite DNA markers.Molecular analyses have shown that populations in the Czech Republic are genetically poor in comparison to foreign populations. At the same time they are entirely different from each other and therefore there is no or very little gene flow between populations. Particularly in populations in the Czech Republic, vegetative reproduction plays an important role, allowing the majority of loci to be fixed in heterozygous form, thus preventing the complete loss of genetic diversity due to genetic drift. Simultaneously, an overwhelming dominance of the single clonal genotype due to competition in each population has reinforced the distinction between geographically close populations. Thus almost every population is a separate genetic line.The vast diversity of populations along with the random allele fixation in populations are probably the remnants of the great genetic diversity of the large Anemone sylvestris population in the past. Its individual parts were fixed by clones.
