Diversity and evolutionary mechanisms of arenaviruses and hantaviruses of small mammals in Eastern Africa

Abstract:

Arenaviry a hantaviry jsou obalené jednořetězcové RNA viry, jejichž přirozenými hostiteli jsou hlodavci a v menší míře též hmyzožravci či letouni. Někteří zástupci jsou významnými patogeny člověka a vyvolávají řadu zdravotních problémů, z nichž část může končit smrtí pacienta. Data o diverzitě a distribuci těchto virů jsou často limitovaná, což platí především pro většinu oblastí subsaharské Afriky, kde mají hlodavci navíc zásadní význam pro veřejné zdraví a kde je značné riziko nákazy pro člověka. V průběhu studie jsme zpracovali kolekci více než 3000 vzorků drobných savců z různých oblastí východní Afriky (Mozambik, Tanzanie, Keňa, Zambie, Etiopie a Demokratická republika Kongo) s cílem zjistit přítomnost i diverzitu arenavirů a hantavirů. Genetickou analýzou byl potvrzen výskyt arenavirů Luna a Morogoro u hlodavce Mastomys natalensis a také detekována přítomnost nových linií arenavirů Mopeia a Mobala. Arenavirus Mobala byl identifikován u pěti různých hostitelských druhů hlodavců tribu Praomyini, což nabourává dřívější teorie o úzké hostitelské specificitě arenavirů. V Keni byla nalezena nová linie již dříve popsaného hantaviru Sangassou u hlodavce Hylomyscus endorobae. Další významný nález pochází z Etiopie, kde byl popsán pro vědu dosud neznámý hantavirus, přenášený zejména abundantním endemickým hlodavcem Stenocephalemys albipes. Virus byl pojmenován Tigray, dle stejnojmenného regionu v severní Etiopii.

Abstract:

Arenaviruses and hantaviruses are enveloped single-stranded RNA viruses, whose primary hosts are rodents and to a lesser extent shrews and bats. Some representatives are important pathogens of humans and may cause serious health problems, from which a part can lead to death of the patient. The data about the diversity and distribution of these viruses are often limited, especially for most areas of the sub-Saharan Africa, where rodents have a major public health relevance and the risk of human infection is considerable. In this study we screened a collection of more than 3000 samples of small mammals from six different countries of Eastern Africa (Mozambique, Tanzania, Kenya, Zambia, Ethiopia and Democratic Republic of the Congo) to investigate the presence and diversity of hantaviruses and arenaviruses. Genetic analysis confirmed the presence of arenaviruses Luna and Morogoro in Mastomys natalensis and also detected new strains of Mobala and Mopeia arenaviruses. Mobala arenavirus was identified in 5 different rodent species from Praomyini tribe, which undermines the previous theory of close host specificity of arenaviruses. The new strain of previously recognized Sangassou hantavirus was found in Hylomyscus endorobae in Kenya. Another important finding comes from Ethiopia, where we described a novel hantavirus, transmitted mainly by very abundant endemic rodent Stenocephalemys albipes. Unknown virus was designated Tigray, according to the same region in northern Ethiopia.
