Abstract:

Práce "Aplikace programu AutoDock 3.0 pro reprodukci rentgenových krystalografických sruktur komplexu acetylcholinesterázy s jejími reaktivátory" se zabývá problematikou užití výpočetních metod při hledání nových reaktivátorů lidského enzymu acetylchlinesterázy.Tyto reaktivátory slouží jako kauzální antidota při otravě nervově paralytickou látkou. Práce se skládá ze dvou částí. První část teoreticky pojednává o metodice úvodních výpočtů struktury komplexu acetylcholinesteráza a jejích reaktivátorů. Součástí teoretické části práce je i popis funkce a významu acetylcholinesterázy při přenosu nervového vzruchu a biochemického fenoménu reaktivace. Ve druhé praktické části je použit program AutoDock 3.0 pro reprodukci RTG struktur komplexů. Konkrétně byl proveden dokovací výpočet struktur s reaktivátory obidoxim, HI6 a HLö-7. Pro každý reaktivátor bylo provedeno pět samostatných výpočtů. Na základě příslušné literatury k dané problematice byl zvolen Lamarkův genetický algoritmus. Zvláštní pozornost byla věnována i zhodnocení relevantnosti řešení vygenerovaných výpočtem. Na závěr byly porovnány struktury získané výpočtem se strukturami získanými RTG difrakcí.

Abstract:

The Final thesis on topic „Application of the AutoDock 3.0 Software for Reproduction of X-ray Crystallographic Structures of Acetylcholinesterase with its Reactivators“ handles with question of using computing methods in searching for new reactivators of human enzyme acetylcholiesterase. These reactivators work as causal antidotes in case of poisoning with nerve agent.This work consists of two parts. First part deal theoretically the methodology of initiative calculations of acetylcholinesterase complex structure and its reactivators. Important part of the theoretical chapter is also description of function and significance of acetylcholiesterase in transfer of neural excitement. In practical part of my work AutoDock software for reproduction of X-ray structures of the complex was used. Calculation of structures was conducted with obidoxim-, HI6- and HLö-7- reactivators. For every reactivator five independent calculations were conducted. On the basis of scientific literature on the topic Lamarckian genetic algorithm have been chosen. Attention have been paid also to the relevance of the solutions generated by the calculations. Finally structures developed through calculation were compared with X-ray structures.
