New trends in methodologies for structural analysis of enzymes

Abstract:

Strukturní biologie makromolekul je věda zkoumající trojrozměrnou strukturu nukleových kyselin a proteinů ideálně na úrovni atomárního rozlišení. Rentgenová krystalografie a nukleární magnetické rezonance jsou již na tomto poli dobře zaběhlými metodami, ale mají svá omezení ať už kvůli složitému a časově náročnému získávání kvalitních biokrystalů, anebo kvůli limitaci ve velikosti biologických makromolekul. Znalost struktury proteinů je klíčová pro pochopení jejich funkce, a proto jsou vyvíjeny nové metody jako kryo-elektronová mikroskopie, sériová femtosekundová krystalografie nebo rozptyl paprsků pod malým úhlem (SAXS), aby doplnily metody stávající a pomohly vyřešit struktury obtížně krystalizovatelných nebo příliš velkých, často konformačně bohatých, proteinových komplexů. Tato práce popisuje tyto nové metody a trendy, které se ve strukturní biologii proteinů, a zvláště enzymů objevují.

Abstract:

Structural biology of macromolecules is scientific discipline that investigates the three-dimensional structures of nucleic acids and proteins, ideally at atomic resolution level. X-ray crystallography and nuclear magnetic resonance are well known and widely used methods, but they still have some limitations such as difficulties with obtaining well-diffracting biocrystals or molecular weight limits. The knowledge on macromolecular structures is fundamental for understanding their biological functions. Therefore, new methods such as cryo-electron microscopy, serial femtosecond crystallography or small-angle light scattering (SAXS) are being continuously developed and improved. These methods can help structurally characterize large, very often conformationally flexible macromolecular complexes, which are difficult to be crystallized. This thesis aims at describing new methods and latest trends of integrative structural biology with emphasis on enzymology.
