Composition of photosynthetic pigments in pigment-protein complexes from barley and chlorina f2 mutant

Abstract:

V této práci jsou nejprve stručně popsány pigmenty v pigment-proteinových komplexech ječmene jarního (Hordeum vulgare L) a jeho mutantu Chloriny f2. Mým cílem bylo stanovit pigmenty v pigment-proteinových komplexech uvedených rostlin užitím elektro-eluce. Nejprve byly metodou elektroforézy získány ze suspenze thylakoidních membrán ječmene jarního (Hordeum vulgare L) separované pigment ? proteinové komplexy v PAGE-gelu. Použitím elektro-eluce se pigment-proteinové komplexy vymyly z PAGE-gelu, přes fritu, která propouští pouze pigment-proteinové komplexy do membránových víček. Aby byly získány pouze pigmenty z pigment-proteinových komplexů, byl přidán aceton, jehož působením proteiny degradovaly. Následnou filtrací byly odstraněny ze vzorku degradované proteiny, čímž byly získány volné pigmenty na analýzu. Analýza pigmentů byla provedena metodou vysokotlaké kapalinové chromatografie. Z výsledného chromatogramu bylo určeno zastoupení jednotlivých pigmentů v pigment ? proteinových komplexech. Elektro-eluce byla provedena na rostlině ječmene jarního (Hordeum vulgare L) pěstovaného při nízké ozářenosti 50 ?mol . Po analýze eluentů pomocí kapalinové chromatografie bylo porovnáno zastoupení jednotlivých pigmentů v určitých pigment-proteinových komplexech.

Abstract:

At first in this bachelor thesis there are briefly described pigments in pigment-protein complexes of barley (Hordeum vulgare L.) and his mutant a Chlorina f2. The principal aim of my work was the estimation of pigments in pigment-protein complexes mentioned plants using an electro-elution. Initially we had to separate pigment-protein complexes in PAGE gel from a suspension of thylakoide membranes from barely (Hordeum vulgare L.). Pigment-protein complexes are washed out from PAGE gel using the electro-elution. Pigment-protein complexes went through a frit which leaked just complexes to a membrane cups. The acetone causes degradation of proteins so we added it for obtaining of the only pigments from pigment ? protein complexes. Free pigments for an analysis were obtained by filtration. The filtration enable take off degraded proteins from the eluent. The analysis of pigments were carried out by usig a high performance liquid chromatography. The representation of the single pigments from pigment ? protein complexes was determined from a resulting chromatogram. The electro-elution method was using for analysing pigments obtained in plants of barely (Hordeum vulgare L.) which were grown under the low irradiance 50 ?mol . After the analysis of eluents by HPLC chromatography we compared the representation of single pigments in pigment-protein complexes.
