The use of lipid analysis for determination of effect of wastes on soil microbial communities

Abstract:

Analýza fosfolipidových mastných kyselin (PLFA) umožňuje odhad kvantity a částečně i kvality půdních mikrobních společenstev. Analýza zahrnuje čtyři hlavní kroky: extrakci celkových lipidů, separaci lipidových frakcí, transesterifikaci fosfolipidů za vzniku methylesterů mastných kyselin (FAME) a analýzu plynovou chromatografií. Na Fakultě životního prostředí UJEP nebyla dosud tato metoda použita. Cílem práce bylo optimalizovat některé kroky této metody a ověřit celý postup na modelovém laboratorním experimentu vlivu těžkých kovů na obsah PLFA lesní půdy. Při optimalizaci bylo zjištěno, že nejvhodnější teplota pro přípravu methylesterů je 37°C a optimální čas 30-45 minut. Nejvyšší výtěžnosti methylesterů při závěrečné extrakci připravených methylesterů pro chromatografickou analýzu bylo dosaženo trojnásobnou extrakcí hexanem. Plynová chromatografie směsného standardu methylesterů mastných kyselin prokázala lineární vztah mezi počtem uhlíků a retenčními časy u homologický řad. Podobnost retenční časů v některých homologických řadách (např. nerozvětvených kyselin a iso-kyselin) nicméně komplikovala identifikaci dle retenčních časů a bylo proto nutné potvrzení identifikace pomocí hmotnostních spekter. Plynová chromatografie také ve vzorcích methylesterů odhalila četné kontaminace pocházející pravděpodobně z pracovního postupu, zejména siloxany a ftaláty. Experiment zaměřený na skladování půdy odhalil největší zastoupení PLFA ve vzorku skladovaném při laboratorní teplotě. V ověřovacím experimentu byl převážně potvrzen negativní vliv těžkých kovů na obsah PLFA i půdní respiraci. Nicméně, zjištěné obsahy PLFA byly nízké a pohybovaly se spíše u meze stanovitelnosti. To naznačuje, že bude nutné optimalizovat ještě další kroky analýzy.

Abstract:

Analysis of phospholipid fatty acids (PLFA) provides an estimate of the quantity and quality of soil microbial communities. The analysis involves four main steps: extraction of total lipids, separation of lipid fractions, transesterification of phospholipids to fatty acid methyl esters (FAME), and analysis by gas chromatography. This method has not been used on the Faculty of Environmental Jan Evangelista Purkyně University in Ústí nad Labem yet. The aim of this work was to optimize certain steps of this method and verify the entire procedure on a model laboratory experiment following the effect of heavy metals on the PLFA amount in forest soil. It was found that the most appropriate temperature for the preparation of FAMEs is 37 °C and the optimal time is 30-45 minutes. The highest yield of methyl esters in the final extraction for chromatographic analysis was achieved with a triple extraction by hexane. Gas chromatography of mixed FAME standard showed a linear relationship between the number of carbons and the retention times in homologous series. The similarity of retention times in some homologous series (e.g. non-branched acids and iso-acids) however complicated the identification by retention times only and confirmation by mass spectra was necessary. Gas chromatography of methyl esters in the samples also revealed number of contamination likely derived from the extraction procedure, especially polysiloxanes and phthalates. The experiment focused on the storage of soil revealed the largest amount of PLFA in the sample stored at room temperature. The verification experiment generally confirmed the negative effect of heavy metals on the PLFA content as well as soil respiration. Identified PLFA contents were however low and were near to quantification limit. This suggests that further optimization steps are necessary.
