Preparation and Study of the Biological Activity of Carbon Quantum Dots

Abstract:

Tato bakalářská práce se zabývá využitím kvantových teček jako nosičů cytostaticky ak-tivních sloučenin. V rámci experimentální části byly připraveny metodou ?bottom up? dva typy uhlíkových kvantových teček. První typ kvantových teček byl připraven pyrolý-zou kyseliny citrónové, druhý typ byl připraven pyrolýzou kyseliny citrónové spolu s glycinem. Dále byly připraveny kvantové tečky obsahující biologicky aktivní látku komplex chlorid chloro(diethtylentriamino)měďnatý. K charakterizaci byly využity metody EPR spek-troskopie, infračervené spektroskopie, optické emisní spektrometrie s indukčně vázanou plazmou a také elementární analýza. Testování cytotoxicity vybraných sloučenin bylo prove-deno na buněčné linii odvozené od akutní T-lymfoblastické leukemie MOLT-4 pomocí WST-1 testu.

Abstract:

This bachelor thesis deals with the use of quantum dots as a carrier of cytostatically active compounds. In the experimental part were prepared two types of carbon quantum dots by ?bottom up? method. First type of quantum dots was prepared by pyrolysis of citric acid and second type was prepared by pyrolysis of citric acid together with glycine. Furthermore, there were prepared carbon quantum dots which contain biological active compound chlo-ro(diethylentriamine)copper(II) chloride (CuDETA). For characterization was used methods of EPR spectroscopy, infrared spectroscopy, optical emission spectrometry with inductively coupled plasma and elementary analysis. Cytotoxicity testing was performed on a cell line derived from acute T-lymphoblastic leukemia called MOLT-4 by use the WST-1 assay.
