Study of the c-myc gene structure in linear and supercoiled DNA

Abstract:

Cílem této diplomové práce bylo studium struktury promotoru c-myc genu v lineární a superhelikální DNA. Toto studium bylo prováděno pomocí chemických a enzymatických strukturních sond. Dále byl posuzován vliv jednomocných a dvoumocných kationtů na vznik lokálních otevřených struktur a také byl studován vliv pH na vznik těchto struktur. Při této práci byly použity plasmidy pBluescript (pBS) a pBS-JW1, kdy je do pBS vložena sekvence o délce 128 bp, které obsahuje oblast hypersenzitivní na nukleázu, tzv. NHE oblast. Jelikož tato oblast obsahuje 6 guaninových repeticí, lze předpokládat vznik lokálních otevřených struktur, v tomto případě kvadruplexů. Proto byl použit právě tento plasmid. Vznikem lokálních otevřených struktur dochází k přeměně stabilní B-DNA formy na non-B DNA formu, která již může být nestabilní. V důsledku přechodu na nestabilní formu DNA může docházet ke zvyšování nebo snižování genové exprese, což se může podílet na mutaci protoonkogenu v onkogennní formy a tím také na procesu kancerogeneze.

Abstract:

The aim of this thesis was to study the structure of the promoter c-myc gene in linear and superhelical DNA. This study was carried out using chemical and enzymatic structural probes. Was also evaluated the effect of monovalent and divalent cations on the formation of local open structures and the influence of pH on the formation of these structures. In this work were used plasmids pBluescript (pBS) and pBS-JW1 when inserted into the pBS sequence with a length of 128 bp, which contains the region hypersensitive to nuclease, called NHE area. Since this region contains six guanine repetition can be expected that local open structures, quadruplexes in this case, so it was just used this plasmid. The creation of local open structures is converted stable B-DNA form of DNA to non-B form of DNA that may already be unstable. As a result of the transition to an unstable form of DNA can result in increase or decrease gene expression, which may contribute to a mutation in the protooncogene to oncogenic form and thus the process of carcinogenesis.
