Significance of cell tests in the diagnosis of allergy

Abstract:

Alergie je přehnaná, nepřiměřená reakce organismu spouštěná podněty, které u většiny jedinců nezpůsobují patofysiologické projevy. Základem alergických onemocnění jsou geneticky podmíněné změny imunologických reakčních mechanismů. Poznání specifické imunologické senzibilizace, určení vlivů prostředí a znalost životního stylu pacientů je cestou k úspěšné léčbě a prevenci. Laboratorní vyšetření zaujímají v diagnostice alergických onemocnění významné místo. Nejvíce jsou využívány testy stanovující alergen-specifické IgE v séru nebo v plasmě a buněčné biologické testy, především pak testy stanovující aktivací basofilů periferní krve. Basotest je jedním z těchto in vitro biologických buněčných testů používaných v diagnostice reakce okamžité přecitlivělosti (alergie) využívající průtokové cytometrie k detekci exprese CD63 molekul na povchu basofilů po stimulaci alergeny pomocí monoklonálních protilátek. Na tento test aktivace bazofilů jsem se zaměřila i ve své diplomové práci.

Abstract:

Allergy is an overreaction of the body triggered by stimuli, that don?t cause pathophysiological response in normal individuals. Genetically dependent changes of the immunologic regulations are the background of the allergy. To identify specific senzitisation, recognition of the enviroment influence and the knowledge of a family life style is the way to successful treatment and prevention. Laboratory diagnostics takes the important place in allergic diseases diagnosis. The most frequently used tests are the tests for the alergen-specific IgE determination and cell biological tests, ie mainly the basophil activation test. Basotest is one of these tests using the flow cytometric determination of CD63 expression on basophiles after alergen stimulation using monoclonal antibodies.
