Modified cucurbit[n]urils: Synthesis and supramolecular interactions

Abstract:

Nedávný rozvoj chemie cucurbit[n]urilů (CB[n], n = 5-10) nás inspiroval, abychom také my přispěli k rozvoji tohoto rychle se rozvíjejícího tématu. První projekt této disertační práce se zabývá vývojem syntézy, oddělování a čištění monosubstituované sloučeniny CB[6], která nese substitutent na pozici methylenové můstku. Metoda zahrnuje reakci glycolurilového stavebního bloku se směsí dvou různých aldehydů - monoaldehydu (RCHO) a formaldehydu. Tuto metodu jsme použili pro přípravu řady monosubstiuovaných CB[6] derivátů vycházejících z různých monoaldehydů jako jsou 3-fenylpropionaldehyd, acetaldehyd, butyraldehyd atd. Surová reakční směs obsahovala velmi složitou směs látek, z tohoto důvodu byla použita hmotnostní spektroskopie MALDI-TOF jako hlavní analytická metoda, pro ověření vzniku substituovaných CB[n] sloučenin. Druhý projekt je věnován syntéze a studiu supramolekulárních vlastností vodorozpustného makrocyklického hostitele, hexamethylcucurbit[6]urilu (MeCB[6]). Studovali jsme vazebné možnosti MeCB[6] s různými kladně nabitými organickými hosty G1 - G4 v roztoku a pevné fázi pomocí 1H NMR spektroskopie a rentgenové difraktometrie. Stanovili jsme asociační konstanty pro komplexy MeCB[6]•G1, MeCB[6]•G3 a MeCB[6]•G4 v čisté vodě pomocí 1H NMR a UV-Vis titrací. Dále jsme určili vliv přítomnosti NaCl ve vodě na tvorbu těchto komplexů.

Abstract:

The recent development outcome of cucurbit[n]uril (CB[n], n = 5-10) chemistry inspired us to contribute efforts to this rapidly growing topic. First project of this dissertation deals with the development of methodology of the synthesis, separation and purification of monosubstituted CB[6] compounds having a substitutent at the methylene bridge position. The method involves the reaction of glycoluril building block with mixture of two different aldehydes - a monoaldehyde (RCHO) and formaldehyde. We used this method for the preparation of a series of mono(substituted) CB[6] derivatives from different monoaldehydes such as 3-phenylpropionaldehyde, acetaldehyde, butyraldehyde etc. Due to complexity of the crude reaction mixture, MALDI-TOF mass spectroscopy was used as the key technique to ascertain the formation of the substituted CB[n] compounds. Second project is dedicated to the synthesis and investigation of supramolecular properties of a water soluble macrocyclic host, hexamethylcucurbit[6]uril (MeCB[6]). We studied the binding modes of MeCB[6] with various organic cationic guests G1 - G4 in the solution and solid state using 1H NMR spectroscopy and X-ray diffractometry respectively. We determined the association constant values of MeCB[6]•G1, MeCB[6]•G3 and MeCB[6]•G4 complexes in pure water using 1H NMR spectroscopic and UV-vis spectrophotometric titration methods. We also estimated the effect of the presence of NaCl in water on the formation of these complexes.
