Genotyping of bacteriocins by food bacteria

Abstract:

Bakteriociny jsou peptidy s antimikrobiální aktivitou syntetizované některými bakteriemi prostřednictvím ribozomů. Bakteriociny dokážou svou schopností ovlivňovat kvalitu a bezpečnost potravin, proto mají vynikající potenciál pro použití jako přírodní konzervační látky v potravinářském průmyslu. Cílem bakalářské práce byla genová typizace bakteriocinů u potravinářsky významných druhů bakterií, konkrétně bakterií mléčného kvašení. V teoretické části je popsána charakteristika, dělení a mechanismus účinku bakteriocinů. Dále je zde zmíněna aplikace bakteriocinů v potravinářství. V práci byly popsány i metody, kterými lze sledovat antimikrobiální aktivita bakteriocinů. V experimentální části byly ověřeny antimikrobiální účinky vybraných sbírkových kmenů (CCDM) bakterií mléčného kvašení pomocí kultivačních metod. U bakteriálních druhů, které prokázaly schopnost inhibice růstu testovaných bakterií, byla provedena izolace a purifikace DNA a genotypizace bakteriocinů metodou PCR. Geny byly vizualizovány prostřednictvím elektroforézy. Za pozitivní producenty bakteriocinů identifikované v této práci lze považovat zástupce rodů Lactobacillus, Lactiplantibacillus, Lactilactobacillus a Lacticaseibacillus.

Abstract:

Bacteriocins are peptides with antimicrobial activity synthesized by some bacteria via ribosomes. Bacteriocins have the ability to influence the quality and safety of food and therefore have excellent potential for use as natural preservatives in the food industry. The aim of the bachelor thesis was the gene typing of bacteriocins in food important bacterial species, specifically lactic acid bacteria. In the theoretical part, the characteristics, division and mechanism of action of bacteriocins are described. Furthermore, the application of bacteriocins to the food industry is mentioned. Methods by which the antimicrobial activity of bacteriocins can be monitored, have also been described in the thesis. In the experimental part, the antimicrobial effects of selected collection strains (CCDM) of lactic acid bacteria were verified by culture methods. The bacterial species that showed the ability to inhibit the growth of the tested bacteria were subjected to DNA isolation and purification and genotyping of bacteriocins by PCR. Genes were visualized by electrophoresis. The positive producers of bacteriocins identified in this work can be considered representatives of the genera Lactobacillus, Lactiplantibacillus, Lactilactobacillus a Lacticaseibacillus.
