The study of plant sources of benzophenanthridine alkaloids

Abstract:

Benzofenanthridinové alkaloidy jsou látky s významnými biologickými účinky. Kromě komerčně dostupných benzofenanthridinů sanguinarinu a chelerythrinu jsou známé také další strukturní typy těchto látek, jako je chelirubin, sanguirubin, chelilutin, sanguilutin a makarpin. Tato práce je zaměřena na podrobnější studium rostlinných druhů, které tyto alkaloidy akumulují. Jde o druhy Macleaya microcarpa a Stylophorum lasiocarpum (Papaveraceae), které jako jediné obsahují ojedinělý a mimořádně biologicky aktivní alkaloid makarpin. Extrakty s obsahem sanguinarinu a chelerythrinu jsou využívány po staletí v lidové medicíně také pro své protizánětlivé účinky, ale mechanismus působení zatím není do detailů znám. Další část disertační práce studuje schopnost alkaloidů sanguinarinu a chelerythrinu ovlivnit genetickou expresi vybraných pro- a protizánětlivých mediátorů.

Abstract:

Benzophenanthridine alkaloids exhibit interesting biological properties. Sanguinarine and chelerythrine are commercially available, but there are also other types of these alkaloids, such as chelirubine, sanguirubine, chelilutine, sanguilutine and macarpine. This thesis is focused on a more detailed study of species Macleaya microcarpa and Stylophorum lasiocarpum (Papaveraceae), that accumulate these alkaloids and that are the only sources of rare and extraordinarily biologically active alkaloid macarpine. Plants with the content of sanguinarine and chelerythrine have been used for centuries in folk medicine for their anti-inflammatory properties, but the mechanism is not fully understood yet. Another part of this thesis is focused on affection of gene expression of inflammatory-related cytokines at the transcription level.
