Molecular and proteomic analysis of lysogenized Staphylococcus aureus strains

Abstract:

Staphylococcus aureus zaujímá vůči svému teplokrevnému hostiteli různé role – může být neškodným komenzálem i nebezpečným patogenem. Příčinou je velký genetický rezervoár, kterým S. aureus disponuje, a jehož důležitou součást tvoří mobilní genetické elementy. Mezi ně se řadí i profágy, které vznikají začleněním genomu mírného bakteriofága do bakteriálního chromozomu v procesu označovaném jako lyzogenní konverze. Zisk genetické informace je doprovázen změnami v genotypu, které se promítají ve fenotypu. Konkrétně při lyzogenizaci ETA-konvertujícími bakteriofágy získá lyzogen schopnost tvořit exfoliatin A, toxin způsobující tvorbu puchýřů a odlupování pokožky. V této práci byla provedena analýza genotypových a fenotypových změn u meticilin rezistentních kmenů S. aureus lyzogenizovaných ETA-konvertujícími bakteriofágy. Byly porovnány různé metody (RPLA, MALDI-TOF profilování, SDS-PAGE, LC-MS/MS) podle jejich schopnosti rozlišení fenotypu původních kmenů a lyzogenů. Tato práce přispívá jednak ke zdokonalení metodiky pro studium sekretomu S. aureus, a zároveň k objasnění vlivu profágů na virulenci a životaschopnost bakterií.

Abstract:

Staphylococcus aureus assumes various roles towards its endothermic host - it can be harmless commensal or dangerous pathogen. This is caused by a vast genetic pool of S. aureus, which is so extensive thanks to mobile genetic elements. One of them – prophages - arise in an event called lysogenic conversion when genome of temperate bacteriophage integrates into bacterial chromosome. The gain of genetic information is followed by changes in genotype, which are projected into phenotype. Specifically, after lysogenization by ETA-converting bacteriophages, the lysogen acquires the ability to produce exfoliative toxin A, a causative agent of blister formation and peeling of skin. In this thesis, I analyzed changes in genotype and phenotype caused by lysogenisation of methicillin-resistant strains of S. aureus by ETA-converting bacteriophages. Several methods (RPLA, MALDI-TOF profiling, SDS-PAGE, LC-MS/MS) were compared according to their ability to differentiate between the original strains and lysogens. This thesis contributes to the improvement of methods used for studying secretome of S. aureus, and to clarification of the impact of prophages on bacterial virulence and fitness.
