Abstract:

Kromě vlastních elektrochemických signálů DNA je v některých případech vhodné využít jejího značení elektroaktivními značkami. Ty zvyšují především specifitu dané analýzy. Takto lze elektrochemickými analytickými metodami studovat strukturu, poškození nebo hybridizace DNA. Jednou z nejdéle používaných značek jsou komplexy oxidu osmičelého s terciálními aminy (Os,L), které tvoří s pyrimidinovými bázemi DNA stabilní, elektroaktivní adukty. Často používané ligandy DNA jsou například 2,2´-bipyridin, 1,10-fenantrolin a N,N,N´,N´-TEMED. Volbou ligandu lze výrazně ovlivnit elektrochemické vlastnosti vznikajících aduktů. To může zvýšit selektivitu a citlivost jejich stanovení. V této práci bude testováno použití 5 nových ligandů na bázi fenantrolinu a chinolinu. Z výsledků analýz vzorků vyplývá, že pouze dva z pěti námi zkoumaných ligandů jsou schopné vytvořit stabilní adukt. Tato skutečnost byla zjištěna na základě analýz reakčních směsí elektrochemickými metodami a dále pak pomocí hmotnostní spektrometrie a kapilární elektroforézy. Reakční podmínky pro oba vybrané ligandy byly optimalizovány pomocí experimentů s krátkým syntetickým oligonukleotidem. Získané výsledky pak byly aplikovány při značení chromozomální DNA izolované z telecího brzlíku.

Abstract:

Besides own electrochemical signals of DNA, in some cases is suitable of use electroactive label brands for its marking. These brands increase primatily the specifity of the analysis. This allows to apply electrochemical analytical methods to study the structure, damage or DNA hybridization. One of the longest used brands are complexes of osmium tetroxide with tertiary amines (Os,L), which form with pyrimidine bases of DNA stable, electroactive adducts. Frequently used DNA ligands are for example 2,2´-bipyridine, 1,10-phenantroline and N,N,N´,N´-TEMED. The ligand choice can significantly affect the electrochemical properties of the nascent adducts. This may increase the selectivity and sensitivity of their determination. In this thesis will be test using five new ligands based on phenanthroline and quinoline. The results of analysis of samples showed us that only two of the five investigated ligands are capable of forming a stable structure. This finding has been based on analyses of the reaction mixtures by electrochemical methods, and then using mass spektrometry and capillary electrophoresis. The reaction conditions for both selected ligands were optimalized through experiments with short synthetic oligonucleotides. The result were applied to marking chromosomal DNA isolated from calf thymus.
