Inovation of separation methods for A\recke{beta} peptides, markers of Alzheimer disease

Abstract:

Pro separaci A\recke{beta} peptidů lišících se o několik aminokyselin bylo vyzkoušeno několik variant a modifikací elektroforetických metod. Jako nejvhodnější metoda pro separaci těchto peptidů byla vybrána Tricin/Tris SDS PAGE elektroforéza s 8 M ureou. Platnost metody byla ověřována na vzorcích reálných sér obohacených o syntetický A\recke{beta} peptid. Dále byla stanovena citlivost stanovení A\recke{beta} peptidu po vizualizaci gelu dusičnanem stříbrným. Separované A\recke{beta} peptidy na Tricin/Tris SDS PAGE elektroforéze s 8 M ureou byly prokázány a vizualizovány pomocí myších specifických protilátek proti N-terminálnímu konci 1 - 17 A\recke{beta} peptidu metodou imunoblottingu. Vizualizace touto metodou byla ověřena na vzorcích reálných sér obohacených o syntetický A\recke{beta} peptid.

Abstract:

Several variants and modifications of electrophoteric method were tested for distingushing among amyloid-\recke{beta} peptides isoforms. As the most suitable the Tricine/Tris SDS PAGE system with 8 M urea added into the gel was chosen. The validity of method was confirmed by analyses of real serum samples enriched by synthetic A\recke{beta} peptide. Furthermore, the sensitivity of assay was determined after vizualization of gel by silver nitrate staining. A\recke{beta} peptides separated on Tricine/Tris SDS PAGE with 8 M urea were manifested and vizualized by means of specific mouse antibodies against N-terminal 1 - 17 domain of A\recke{beta} peptide through the use of immunoblotting technique. This vizualization was confirmed again by real serum samples enriched by synthetic A\recke{beta} peptide.
