Thermodynamic Dissociation Constants of Selected Cytostatics and Immunosuppressives

Abstract:

Jsou stanoveny termodynamické disociační konstanty imunosupresiv a cytostatik azathioprinu, methotrexátu, rasagilinu báze, rasagilinu mesylátu a cyklosporinu A (IVAX Pharmacueticals s.r.o., Opava) technikou pH-spektrofotometrické titrace při 25°C a 37°C. Po stanovení počtu ve spektrech indikovatelných světlo-absorbujících částic algoritmem INDICES byla ze závislosti smíšených disociačních konstant na odmocnině z iontové síly nelineární regresní analýzou zkráceného Debye-Hückelova vztahu programem ADSTAT odhadnuta termodynamická disociační konstanta. Nalezené odhady byly v dobré shodě s predikovanými odhady strukturní analýzy kvantově-chemického programu PALLAS.

Abstract:

Thermodynamic dissociation constants of cytostatics and immunosuppressants azathioprine, methotrexate, rasagiline base, rasagiline mesylate and cyclosporine A (IVAX Pharmacueticals s.r.o.) were estimated by technique of pH-spectrophotometric titration at 25°C and 3°C. The first step was the determination in the spectrum indicated particles by the algorithm INDICES, then was from a dependence of dissociation constants on the root of the ionic strength by non-linear regression analysis of Dębye ? Hückelov law with the programme ADSTAT estimated the thermodynamic dissociation constant. Found estimates of pKaT were in good agreement with predicted estimates of structural analysis quantum-chemical programme PALLAS.
