Toxicity and oxidative stress responses in plants exposed to polycyclic aromatic compounds adn their N-heterocyclic derivates

Abstract:

Polycyklické aromatické uhlovodíky a jejich N-heterocyklické deriváty (NPAHs) jaou v přírodě široce rozšířené, byly detekovány ve vzduchu, vodě, sedimentech a půdě. Ačkoli je známo, že jsou vysoce toxické pro některé organismy, úplné porozumění jejich environmentálního osudu, biologického metabolismu a účinků je omezené. V rámci studie byly zjištěny toxické efekty tří homocyklických aromatických uhlovodíků (PAHs - fenantrenu, antracenu, fluorenu) a jejich sedmi N-heterocyklických derivátů (fenantridinu, 1,10-fenantrolinu, 1,7-fenantrolinu, 4,7-fenantrolinu, benzo(h)chinolinu, akridinu, karbazolu) na vyšší rostliny Sinapis alba, Triticum aestivum a Phaseolus vulgaris. Byly studovány morfologické endpointy (délka, hmotnost, klíčivost), parametry detoxikace a antioxidativní komponenta rostlinného metabolismu a rovněž lipidní peroxidace v akutním (čtyřdenním) testu fytotoxicity. Toxicita NPAHs byla celkově vyšší než účinky parentálních homocyklických aromatických uhlovodíků a významně se lišila v závislosti na struktuře individuálních NPAHs. Všechny testované chemikálie významně indukovaly aktivitu detoxifikačních a antioxidativních enzymů (glutation reduktázy, glutation peroxidázy, glutation-S-transferázy) v nanomolárních a nízkých mikromolárních koncentracích. Výsledky této studie nově shrnují a ukazují významnou fytotoxicitu NPAHs a ukazují vhodnost použití biomarkerů oxidativního stresu a detoxifikace pro hodnocení toxicity a jako časné varování v případě fytotoxicity in vivo.

Abstract:

Polycyclic aromatic hydrocarbons and their N-heterocyclic derivatives of (NPAHs) are widespread and have been detected in air, water, sediments and soil. Although they were shown to be highly toxic to some organisms, our understanding of their occurrence, environmental fate, biological metabolism and effects is limited. This study evaluated toxic effects of three homocyclic aromatic hydrocarbons (PAHs - phenanthrene, anthracene, fluorene) and their seven N-heterocyclic derivates (phenanthridine, 1,10-phenanthroline, 1,7-phenanthroline, 4,7-phenanthroline, benzo[h]quinoline, acridine, carbazole) on higher plants Sinapis alba, Triticum aestivum and Phaseolus vulgaris. Morphological endpoints (growth, weight, germination), parameters of detoxification and antioxidant components of plant metabolism as well as lipid peroxidation were studied in acute (4 day) phytotoxicity tests. Toxicity of NPAHs was generally more pronounced than the effects of parent PAHs, and it significantly differed with respect to the structure of individual NPAHs. All tested chemicals significantly induced activities of detoxification and antioxidant enzymes (glutathione reductase, glutathione peroxidase, and glutathione-S-transferase) at nanomolar to low micromolar concentrations. The results of our study newly demonstrate significant toxicity of NPAHs to plants and reveal suitability of multiple biomarker assessment to characterize mechanisms of oxidative stress and serve as an early warning of phytotoxicity in vivo.
