Immunohistochemical profile of selected interleukins and their relation to cytochrome P450 epoxygenases in placental Hofbauer cells

Abstract:

Stroma placentárních klků obsahuje četné makrofágy, tzv. Hofbauerovy buňky, které jsou fetálního původu a hrají roli v řadě procesů během gravidity. Pomocí imunohistochemie jsme studovali expresi interleukinů IL-1β, IL-10 a CYP epoxygenáz CYP2C8, CYP2C9, CYP2J2 v Hofbauerových buňkách s následným měřením a porovnáním barevné intenzity v softwaru Fiji. Tkáně placenty byly získány z normálních gravidit v různém gestačním stáří (od 6. do 13. týdne a placenty při porodu). Prokázali jsme expresi interleukinů IL-1β, IL-10 a CYP epoxygenáz CYP2C8, CYP2C9, CYP2J2 v Hofbauerových buňkách placenty v různých časových obdobích gravidity pomocí imunohistochemie. Exprese IL-10 s gestačním věkem narůstala, zatímco exprese IL-1β se chovala zcela opačně. Exprese námi studovaných CYP epoxygenáz vykazovala různou úroveň v závislosti na gestačním věku. Nejvýrazněji byla exprimována CYP2C8 a narůstala s gestačním stářím. V Hofbauerových buňkách jsme dále prokázali pozitivní korelaci exprese CYP2C8 s IL-10 a zároveň negativní korelaci s IL-1β. Tyto skutečnosti potvrzují klíčovou roli Hofbauerových buněk v imunologických pochodech, během různých období gravidity.

Abstract:

The villous stroma of the placenta contains numerous macrophages called Hofbauer cells which are of foetal origin and are believed to function in many processes. Using immunohistochemistry we studied the expression of interleukins IL-1β, IL-10 and CYP epoxygenases CYP2C8, CYP2C9, CYP2J2 in Hofbauer cells, followed by detection of reciprocal intensity in Fiji software. Placental tissues were obtained from normal pregnancies at various gestational age (from 6th to 13th week and term placenta). We demonstrated the expression of interleukins IL-1β, IL-10 and CYP epoxygenases CYP2C8, CYP2C9, CYP2J2 in Hofbauer cells of the placenta at various gestational ages by immunohistochemistry. The expression of IL-10 increased with gestational age, while IL-1β expression behaved quite the opposite. The expression of CYP epoxygenases we studied showed varying levels depending on gestational age. Most notably, CYP2C8 was expressed and increased with gestational age. In Hofbauer cells we also demonstrated a positive correlation of CYP2C8 expression with IL-10 and a negative correlation with IL-1β. These facts confirm the key role of Hofbauer cells in immunological processes, during various periods of pregnancy.
