In vitro evaluation of cytotoxity of antidotes for the treatment of poisoning by organophosphorus compounds from the group of cholinesterase reactivators

Abstract:

Při terapii intoxikace organofosforovými sloučeninami ze skupiny inhibitorů cholinesteras je nejdůležitější léčba reaktivátory acetylcholinesterasy. Z pohledu její účinnosti je velmi důležité, jakou látkou je osoba intoxikována a jaký reaktivátor se použije v léčbě. Proto je snaha syntetizovat nové reaktivátory, které by spojovaly výhody zavedených standardních reaktivátorů, a zároveň by byly účinné vůči širokému spektru inhibitorů. Tato diplomová práce se zabývá problematikou oximových reaktivátorů cholinesteras, sloužících jako antidota při otravě organofosforovými pesticidy nebo bojovými chemickými látkami s nervově paralytickým účinkem. Dále je zaměřena na význam acetylcholinu a cholinesteras v nervovém systému. Součástí druhé části je úvod do základů práce s buněčnými liniemi a následně úvod do hodnocení in vitro cytotoxicity vybraných reaktivátorů cholinesteras. Experimentální část se zabývá samotným provedením testu pro hodnocení in vitro cytotoxicity.

Abstract:

For the therapy intoxication by organophosphorus compounds from the group of cholinesterase inhibitors is very important treatment with acetylcholinesterase reactivators. Its efficacy is influenced by the substance which caused the poisoning and what reactivator was used. Therefore, there is the effort to synthesize the new oximes, which combines the advantages of established standard reactivators and would also be effective for a wide spectrum of inhibitors. This thesis describes in the first part the oxime cholinesterase reactivators, which are used as antidotes for poisoning by organophosphorus pesticides and chemical warfare nerve agents. There is described the role of acetylcholine and cholinesterases in the nervous system. The second part of this thesis is an introduction to the basics of working with cell lines and an introduction to evaluation of in vitro cytotoxicity of selected cholinesterase reactivators . Experimental deals with in vitro cytotoxicity testing.
