Optimalization of the storage conditions for bovine testicular hyaluronidase

Abstract:

Hyaluronidázy (HYALs) jsou enzymy patřící do skupiny glykosidáz. Jejich funkcí je degradace hyaluronanu (HA), který je jako složka extracelulární matrix (ECM) zapojen do mnoha fyziologických procesů. HYALs jsou tradičně využívány pro regulaci množství HA při medicínských aplikacích a díky své schopnosti zvyšovat propustnost membrán a difuzi léčiva nalezly využití také ve farmakologii. Aktuálním tématem je výzkum role HYALs při růstu maligní tkáně či zapojení HYALs v rámci patogeneze různých onemocnění. V práci byly optimalizovány podmínky pro uchovávání enzymu bovinní testikulární hyaluronidázy (BTH), který je z důvodu vysoké sekvenční podobnosti tradičně využíván pro studium lidských HYALs. Získaná data ukazují, že dlouhodobým uchováním zásobního roztoku enzymu dochází k poklesu aktivity testované BTH, nikoli však k úplné inaktivaci enzymu. Po 48 hodinách uchování je residuální aktivita enzymu vyšší než 80 % pro všechny testované kombinace skladovacích podmínek. Po 336 hodinách zůstává vyšší residuální aktivita u enzymu, který byl skladován v pufru o pH 5,4, a to 67,16 % (při laboratorní teplotě) a 66,42 % (při -80 °C).

Abstract:

Hyaluronidases (HYALs) are enzymes belonging to the family of glycosidases. Their function is the degradation of hyaluronan (HA), which is involved in many physiological processes, being a component of the extracellular matrix (ECM). HYALs are traditionally used to control the HA levels in medical applications and due to their ability to increase membrane permeability and drug diffusion, they are also used in pharmacology. The current topic of the research of the role of HYALs in growth of malignant tissue or the involvement of HYALs in pathogenesis of various diseases is an actual topic. In this work, we studied the optimization of the storage conditions for the bovine testicular hyaluronidase (BTH) enzyme, which is traditionally used to study the human HYALs because of its high sequence similarity. The obtained data show that long-term storage of the enzyme stock solution resulted in decreased activity but not in complete inactivation of BTH enzyme. After 48 hours of storage, the residual enzyme activity was bigger than 80 % for all tested combinations of storage conditions. After 336 hours, the residual activity of the enzyme stored in a buffer of pH 5.4 remained 67.16 % (at laboratory temperature) and 66.42 % (at -80 °C).
