Cytogenetic methods and their application in the study of monogenean karyotypes

Abstract:

Monogenea je skupina převážně rybích parazitů s přímým a jednoduchým, ale unikátním životním cyklem. Cytogenetika monogeneí je poměrně neprobádanou oblastí, která může přinést nové, významné poznatky průběhu evoluce chromozomů těchto druhů a zároveň být nápomocná při řešení fylogenetických vztahů. Tato práce odhaluje pro vědu nová data o chromozomech běžného parazita kaprů Eudiplozoon nipponicum. Karyotyp tohoto druhu sestává ze sedmi párů telocentrických chromozomů (2n=14t, TLC=44,55 um). Na všech chromozomech byly v pericentromerické oblasti po barvení DAPI zřetelné heterochromatinové bloky. Stříbření (AgNO3) a barvení pomocí fluorescenční barvičky YOYO-1 odhalilo přítomnost jednoho velkého, aktivního, jadérka. Fluorescenční in situ hybridizace s 18S rDNA sondou odhalila jeden cluster ribozomálních genů situovaný v terminální části dlouhých ramínek chromozomálního páru č. 7. Výsledky byly v souladu s hypotézou od Korolevy, která tvrdí, že karyotyp s nižším počtem chromozomů je původní a proto, karyotyp E. nipponicum může být považován za ancestrální v čeledi Diplozoidae.

Abstract:

Monogenea is a group of predominantly fish parasites with direct and simple, but unique life cycle. Cytogenetics of monogeneans is a relatively unexplored area, but it can bring new, significant information about the evolution of the chromosomes of these species while helping to solve phylogenetic relationships within the taxa. The present study reveals new data on chromosomes of a common parasite of carps Eudiplozoon nipponicum. The karyotype of this species is composed of seven pairs of telocentric chromosomes (2n=14t, TLC=44,55 µm). Heterochromatin blocks were marked on all chromosomes in the pericentromeric region after DAPI staining. Silver staining (AgNO3) and staining with fluorescent dye YOYO-1 revealed the presence of one large, active, nucleus. Fluorescent in situ hybridization with 18S rDNA probe revealed one cluster of ribosomal genes situated at the terminal part of the long arms in chromosome pair No. 7. The results were consistent with the hypothesis by Koroleva, which claims that the karyotype with the lower number of chromosomes is ancient and thus, E. nipponicum may be considered as the species with ancient karyotype in the family Diplozoidae.
