Abstract:

Cytokininy jsou rostlinné hormony účastnící se mnoha fyziologických a vývojových procesů v rostlině. Přirozenými cytokininy jsou N6-substituované deriváty adeninu, které jsou dle typu substituovaného řetězce děleny na aromatické a isoprenoidní. Cytokininy jsou degradovány v jednokrokové enzymatické reakci, která je katalyzována enzymem cytokinindehydrogenasou (CKX, EC 1.5.99.12). Tento enzym patří mezi klíčové regulátory obsahu aktivních cytokininů v rostlinách a inhibice CKX může mít zajímavý biologický účinek. Tématem této práce bylo provedení CKX inhibičních testů na derivátech anilinopurinu a následné stanovení vlivu těchto látek na jednotlivé zkoumané enzymy AtCKX1, AtCKX2 a ZmCKX1. Dále byla určována IC50 jednotlivých látek a bylo posuzováno, která sloučenina je z testovaných látek významným inhibitorem daných enzymů. Byly určeny nové inhibitory CKX odlišující se navzájem schopností inhibovat dané enzymy z Arabidopsis thaliana a Zea mays.

Abstract:

Cytokinins are plant hormones which participate in many physiological and developmental processes in plants. The natural cytokinins are N6-substituated derivatives of adenine. They can be aromatic or isoprenoid according to the type of substituated chain. Cytokinins are degradated in one-step reaction which is catalysed by cytokinindehydrogenase (CKX, EC 1.5.99.12). Content of the cytokinins in plants is regulated by this key enzyme and thus inhibition of the CKX can have interesting biological effect. The theme of this work was to carry out CKX inhibition tests to determine influence of provided compounds on enzymes AtCKX1, AtCKX2 a ZmCKX1. Then IC50 values were determinated and it was assessed which compound is a better inhibitor of each enzyme. Some of tested compounds has been demonstrated as the good inhibitors of individual enzymes. The new CKX inhibitors differing in the ability to inhibit the enzymes from Arabidopsis thaliana and Zea mays have been identified.
