Abstract:

Polycyklické aromatické uhlovodíky (PAHs) představují svým rozšířením v biosféře, rizikovými vlastnostmi (karcinogenita, mutagenita a teratogenita) a schopností akumulace v živočišných tkáních a rostlinných pletivech riziko pro všechny živé organismy. Rostliny stojící na počátku potravního řetězce umožňují vstup těchto xenobiotických sloučenin do vyšších trofických úrovní. V závislosti na délce a intenzitě působení mohou PAHs vyvolávat akutní, chronické i latentní poškození rostlin. Údaje hodnotící vliv PAHs na biochemické a fyziologické procesy determinující růst i vývoj rostlin jsou doposud nepočetné. Podobně jako PAHs také herbicidy, skupina organických sloučenin úmyslně vnášených do životního prostředí, mohou při intenzivním používání způsobovat kontaminaci půdy, vody, ovzduší i samotných zemědělských plodin. Z půdy a vody se pak také dostávají do rostlinstva a dále do potravního řetězce. Se zvyšujícím se stupněm zatížení životního prostředí toxickými látkami (PAHs, herbicidy) nabývá na aktuálnosti vývoj a uplatnění nových metod pro včasnou indikaci stresu a působení organických sloučenin na organismy. Předložená disertační práce shrnuje doposud známé poznatky o působení polycyklických aromatických uhlovodíků a herbicidů na růst a vývoj rostlin. Zároveň však doplňuje informace o působení těchto organických sloučenin na jednu z nejdůležitějších dílčích reakcí fotosyntézy, Hillovu reakci. Pozornost byla také zaměřena na změny v pigmentoproteinových komplexech při absorpci a přenosu záření a toku elektronů z fotosystému II k fotosystému I. Ke studiu fotochemických procesů na úrovni tylakodní membrány chloroplastů byla použita metoda indukované fluorescence chlorofylu. V rámci práce byl studován vliv zvyšující se koncentrace fluoranthenu (FLT) a fotomodifikovaného FLT (fFLT) (0,01; 0,1, 1 a 5 mg L-1) a herbicidu Basagranu 600 s účinnou látkou bentazon (5, 20, 35 a 50 nmol L-1) na klíčivost a růst klíčních rostlin a fotosyntetické procesy rostlin hrachu (Pisum sativum L., cv. Garde). Významné snížení klíčivosti bylo pozorováno pod vlivem FLT a fFLT již při nízké koncentraci (0,01 mg L-1). Zatímco inhibice klíčivosti rostlin v přítomnosti bentazonu byla zaznamenána až při působení vyšších koncentrací (35 a 50 nmol L-1). Získané výsledky také dokládají, že obě formy FLT způsobily významnou inhibici délky kořene a nadzemní části rostlin až při působení koncentrace 1 mg L-1. Bentazon inhiboval růst kořene i nadzemní části již při koncentraci 5 nmol L-1. Změny parametrů fluorescence chlorofylu a aktivity Hillovy reakce dokládají ovlivnění primárních procesů fotosyntézy. Měření bylo prováděno na suspenzi chloroplastů izolovaných z rostlin hrachu. Významné zvýšení hodnoty F0 a snížení FV/FM a ΦII bylo zaznamenáno při koncentracích 0,1 a 1 mg L-1 FLT a 0,01; 0,1 a 1 mg L-1 fFLT. Významné zvýšení parametru indukované fluorescence chlorofylu F0 a snížení hodnoty FV/FM bylo zaznamenáno až při působení nejvyšší koncentrace (50 nmol L-1) bentazonu. Zatímco hodnota kvantového výtěžku fotosystému II byla inhibována již působením 5 nmol L-1. Mnohem citlivějším ukazatelem kontaminace se ukázala aktivita Hillovy reakce. Zatímco FLT a fFLT významně inhiboval produkci kyslíku vlivem koncentrací 0,01 a 1 mg L-1. Bentazon snížil aktivitu Hillovy reakce již při koncentraci 5 nmol L-1. Až 91% inhibice aktivity Hillovy reakce byla způsobena 50 nmol L-1 bentazonu. Experimenty prokázaly analogické působení PAH fluoranthenu a herbicidu Basagranu 600. Dále bylo prokázáno, že změny v aktivitě Hillovy reakce lze využít jako marker poškození rostlin vlivem životního prostředí.

Abstract:

Polycyclic aromatic hydrocarbons (PAHs) are widely spread compounds in the biosphere. Some properties of these compounds (carcinogenic, mutagenic and teratogenic) as well as their ability to accumulate in tissues represent a risk for all living organisms. Plants standing on the beginning of the food chain enable the xenobiotic compounds to penetrate into the higher trophic levels. Depending on the time and intensity of influence PAHs can evoke acute, chronic and latent injury of plants. Information that allows evaluation of the effects of PAHs on biochemical and physiological processes are scarce. Similarly to PAHs, herbicides, a group of organic compounds intentionally introduced in the environment by humans, may also cause the contamination of soil, water and air as well as crop plants when used to excess. Herbicides may consequently move further in the food chain, too. Rising level of pollution load by toxic compounds like PAHs or herbicides increases an importance of the development and broader use of new methods for early indication of stress due to the exposure to organic contaminants and the effects of contaminants on living organisms in general. This thesis summarizes current knowledge about the effects of PAHs on plant growth and development. Simultaneously, it also brings information about effects of organic contaminants on one of the most important parts of photosynthesis in plants - the Hill reaction. The attention was also aimed to changes in pigment-protein complexes where the light energy is absorbed and transported in electron flux from photosystem II to photosystem I. Photochemical processes at the level of thylakoid membrane were studied using a method of induced chlorophyll fluorescence. In this thesis the effects of increasing concentration of fluoranthene (FLT), photo-modified fluoranthene (phFLT) (0.01, 0.1, 1 and 5 mg L-1) and herbicide Basagran 600 with active compound bentazone (5, 20, 35 and 50 nmol L-1) were studied on germination rate, growth of pea plant seedlings and primary processes of photosynthesis. Significant decrease of the number of germinating seeds was observed even under low concentrations of both FLT and phFLT (0.01 mg L-1). In contrast, the inhibition of germination was observed under higher concentrations of bentazon (35 a 50 nmol L-1). The results also demonstrate that both forms of FLT caused significant reduction of root and shoot length in concentration of 1 mg L-1. Bentazon inhibited root and shoot growth in concentration as low as 5 nmol L-1. Changes in the parameters of induced chlorophyll fluorescence and in Hill reaction activity demonstrated the effect of studied compounds on the primary processes of photosynthesis. Measurements were done on a suspension of chloroplasts isolated from pea plants. Significant increase of basal chlorophyll fluorescence (F0) value and decrease of parameters FV/FM and ΦII was recorded in concentrations 0.1 a 1 mg L-1 FLT and 0.01, 0.1 and 1 mg L-1 phFLT. Significant increase of parameter F0 and decrease of ratio FV/FM was found only in the highest concentration of bentazone (50 nmol L-1). On the other hand, the decrease of the quantum yield of the photosystem II (ΦII) was found already in response to concentration 5 nmol L-1 of bentazone. The activity of the Hill reaction was shown as much more sensitive marker of contamination. FLT and phFLT significantly inhibited oxygen production in concentrations 0.01 and 1 mg L-1. The significant effect of bentazone was recorded already at 5 nmol L-1 and the degree of inhibition reached about 91% at concentration 50 nmol L-1. The results of this thesis demonstrated some analogy in mechanisms of the impact of PAHs and some herbicides. They also showed that Hill reaction activity is a useful and sensitive marker of plant damage caused by environmental contamination.
