Temperature impact on functional state of isolated thylakoid membranes

Abstract:

Cílem této bakalářské práce bylo vypracovat literární rešerši na téma vlivu zvýšených teplot na funkčnost fotosyntetického aparátu vyšších rostlin. Experimentální část se zabývá izolací thylakoidních membrán smrku ztepilého a vyhodnocení vlivu zvýšených teplot na thylakoidní membrány pomocí měření fluorescence chlorofylu a (PAM fluorimetr), a stanovení maximální fotochemické účinnosti PSII. Maximální fotochemická účinnost PSII smrku ztepilého byla pozorována při 32 °C (a 20 °C), a se zvyšující se teplotou dochází ke snižování tohoto parametru. To je ve shodě s literaturou, která uvádí, že při teplotách 35 ? 42 °C dochází k mírnému tepelnému stresu, při kterém dochází ke snížení enzymové kinetiky, a rostliny přebytečnou energii disipují z větší části ve formě tepla. Suspenze thylakoidních membrán smrku ztepilého nevykazovaly typické zhášení fluorescence chlorofylu a při působení vyšší ozářenosti (1400 ?mol m-2 s-1) a zvýšené teploty. Při izolaci thylakoidních membrán zřejmě došlo k vyplavení přenašečů elektronů na straně PSI (ferredoxinu) a neprobíhal, lineární transport elektronů přes PSI a PSII. Zároveň nebyla pozorována konverze violaxantinu na zeaxantin ani v případě thylakoidních membrán smrku obohacených o přídavek askorbátu (30 mM) a úpravě pH na optimální hodnotu (pH = 5) pro funkci violaxantin deepoxidázy. Pro účely měření fluorescence na izolovaných thylakoidních membránách by bylo vhodné do zásobního média (o pH = 5) přidat umělý akceptor elektronů z PSI (např. methylviologen) a askorbát.

Abstract:

The aim of this bachelor thesis was to conclude a literature review on the temperature impact on functional state of photosynthetic apparatus of higher plants. The experimental part deals with the isolation of thylakoid membranes of Norway spruce and the evaluation of the effects of elevated temperatures on the thylakoid membranes by measuring the fluorescence of chlorophyll a (PAM fluorimeter) and the determination of the maximum photochemical efficiency of PSII. The maximum photochemical efficiency of PSII of Norway spruce was observed at 32 °C (and 20 °C) and that with increasing temperature there is a reduction of this parameter. The resultes are consistent with the literature, which presents that at temperature 35 ? 42 °C there is a slight heat stress, which leads to a reduction of enzyme kinetics and plants dissipates excess energy mostly in the form of heat. The suspensions of thylakoid membranes of Norway spruce showed no typical quenching of chlorophyll fluorescence at expose to higher irreadiance (1400 ?mol m-2 s-1) and an increase temperature. The isolation of thylakoid membranes caused wash out of the electron carriers on the side of PSI (ferredoxine) and linear electron transport didn´t take place throught PSI and PSII. At the same time the conversion of violaxanthin to zeaxanthin wasn´t observe even if thylakoid membranes of Norway spruce were enriched in the addition of ascorbate (30 mM) and pH was adjusted to the optimal value (pH = 5) for the function of violaxanthin de-epoxidase. In the isolated thylakoid membranes would be suitable to add an artificial electron acceptor of PSI (such as methylviologen) and ascorbate to the storage medium for fluorescence measurement.
