Elastic surfaces for biomechanical study of cardiomyocytes

Abstract:

Diplomová práce je zaměřena na studium některých elastických povrchů, které jsou potenciálně vhodné pro studium srdečních buněk a jejich biomechanických vlastností a mohly by být využity v medicínské praxi. Práce se zaměřuje na studium biokompatibilních elastických povrchů, které mohou simulovat mechanické vlastnosti srdeční tkáně a mohly by tak zajistit vhodný podklad pro kultivaci kardiomyocytů, které jsou v medicínských laboratořích využívané především ke studiu mnoha srdečních abnormalit a onemocnění. Byly připravovány polyakrylamidové (různý poměr akrylamidu a bisakrylamidu) a hyaluronanové (obsahující i neobsahující želatinu) gely, jako referentní vzorek bylo použito sklo. Příprava těchto vybraných elastických povrchů byla optimalizována a gely byly analyzovány pomocí mikroskopie atomárních sil. Mechanické vlastnosti gelů byly analyzovány metodou nanomechanického mapování a byl studován vliv těchto vlastností na strukturu a mechanické vlastnosti buněk. Ke studiu struktury buněk byla navíc použita metoda barvení aktinových vláken. U buněčného materiálu se podařilo najít souvislost mezi tuhostí hyaluronanových gelů a tuhostí buněk, ale také mezi tuhostí povrchu a výškou buňky kultivované na tomto povrchu. Připravené hyaluronanové gely byly biokompatibilní a svou tuhostí odpovídaly tuhosti buňky.

Abstract:

The presented Master´s thesis is focused on characterization and specification of selected elastic surfaces that would be potentially useful for studying heart cells and their biomechanical properties and which could be used in biomedical engineering. The elastic biocompatible surfaces were studied as they could simulate heart tissue natural properties, and thus would provide a suitable substrate for culturing cardiomyocytes. Those cells are mainly used for the study of many diseases and heart abnormalities in medical laboratories. Polyacrylamide (different ratio of acrylamide and bisacrylamide) and hyaluronate (containing or non-containing gelatin) gels were prepared, glass was used as the reference sample. The preparation of these elastic materials was optimized and gels were analyzed by atomic force microscopy. Mechanical properties of the gels were analyzed by nanomechanical mapping and the effect of these properties on the structure and mechanical properties of the cells was studied. In addition, the method of actin fibers staining was used to analyze the cell structure. As for the cell material, there is a correlation between the stiffness of the hyaluronan surfaces and the stiffness of the cell, but also between the stiffness of the surface and the height of the cell which is cultivated on that surface. These prepared hyaluronate gels were biocompatible and their stiffness correspond with the cell stiffness.
