Function of desaturases and elongases in human lipid metabolismus

Abstract:

Tato bakalářská práce se zabývá funkcí desaturáz a elongáz v metabolismu lipidů člověka. Zkoumá jejich význam v metabolismu endogenních a exogenních mastných kyselin na příkladu laboratorních zvířat i člověka. Jsou zde popsány příčiny poruch, jejich fyziologické projevy a souvislost s výskytem hyperglykemie, dyslipidemie, vznikem inzulinové rezistence následované vznikem metabolického syndromu, diabetu mellitu 2. typu a možných následných kardiovaskulárních chorob. Dále je uvedena používaná vyšetřovací a diagnostická metodika, jejich experimentální dostupnost, účinnost a přesnost. Význam studia desaturáz a elongáz nabývá na důležitosti, moderní genetické metody umožňují podrobnější studium delecí jednotlivých genů na laboratorních zvířatech. Nasycené mastné kyseliny toxicky působí na celý organismus a spolu s oxidačním stresem mohou mít vliv na vznik takzvaných civilizačních chorob.

Abstract:

This bachelor thesis deals with the function of desaturases and elongases in human lipid metabolism. It examines their role in the metabolism of endogenous and exogenous fatty acids on example of laboratory animals and even humans. It describes causes of disorders, their physiological manifestations and association with hyperglycaemia, dyslipidaemia, insulin resistance followed by the emergence of the metabolic syndrome, second type of diabetes mellitus and the possibility of subsequent cardiovascular diseases. There are also mentioned used investigative and diagnostic methods, their experimental accessibility, efficiency and accuracy. The studying of desaturases and elongases is gaining importance, modern genetic techniques allow a more precision study of deletions single genes in laboratory animals. Saturated fatty acids affect the whole organism toxically, and with oxidative stress may influence the formation of so-called lifestyle diseases.
