\kur{In-vitro} Fertilization and Epigenetics

Abstract:

Tato bakalářská práce se zaměřuje na problematiku asistované reprodukce, zejména na IVF a epigenetiku. V úvodní části podrobně popisuje jednotlivé fáze IVF, včetně stimulace vaječníků, preimplantačního genetického testování a kryokonzervace. Následuje vysvětlení epigenetiky, vědního oboru, který studuje dědičné změny v genové expresi bez změn v samotné DNA sekvenci. Mezi hlavní epigenetické mechanismy, které stabilně ovlivňují genomovou aktivitu prostřednictvím mitózy, patří DNA metylace, histonová metylace a RNA interference. Práce se také zabývá genomickým imprintem, což je proces umlčování genů pomocí DNA metylace. V závěrečné části se soustředí na rizika vzniku imprintových chyb při asistované reprodukci a některé nemoci, které z těchto chyb mohou vyplývat.

Abstract:

This bachelor thesis focuses on assisted reproduction, in particular on IVF and epigenetics. In the introductory part it describes in detail the different stages of IVF, including ovarian stimulation, preimplantation genetic testing and cryopreservation. This is followed by an explanation of epigenetics, a scientific field that studies heritable changes in gene expression without changes in the DNA sequence itself. The major epigenetic mechanisms that stably affect genomic activity through mitosis include DNA methylation, histone methylation, and RNA interference. This paper also discusses genomic imprinting, which is the process of gene silencing by DNA methylation. The final section focuses on the risks of imprinting errors in assisted reproduction and some of the diseases that can result from these errors
