Abstract:

Předkládaná dizertační práce se zabývá magnetorecepcí, zvláštní schopností zvířat vnímat magnetické pole Země. Přestože je tato schopnost již půlstoletí intenzivně zkoumána, samotný magnetoreceptor dosud nalezen nebyl, nebyl objasněn ani přesný mechanismus zpracování a u řady živočichů neznáme ani význam recepce magnetické informace. Úvodní část představuje dva uvažované mechanismy magnetorecepce, přičemž větší pozornost je věnována magnetorecepci závislé na světle. Zvláštní důraz je v této práci kladen na schopnosti rozlišit směrový vektor magnetického pole. Práce také reflektuje v současnosti stále více diskutované téma opakovatelnosti získaných dat. Jako součást práce jsou přiloženy tři články, jejichž výsledky jsou prezentovány a diskutovány. První studie sleduje cíl verifikovat a rozšířit znalosti o nejjednodušší směrové odpovědi, magnetickém alignmentu – magnetickým polem podmíněném pozičním chování. Ve zbývajících dvou pracech dokazujeme s použitím odlišných diagnostických přístupů spojení směrové orientace s mechanismem radikálových párů. Ve studii II využíváme metody RNA interference, kdy po vypnutí kryptochromu 2 švábi ztrácí schopnost vnímat změny směru magnetického pole. Studie III ukazuje narušení směrové orientace mořského korýše Gondogeneia antarctica v ose moře-souš po aplikaci slabého radiofrekvenčního pole, což je pokládáno za diagnosticky přesný znak svědčící pro mechanismus radikálových párů.

Abstract:

This dissertation deals with magnetoreception - a remarkable ability of animals to perceive the Earth's magnetic field. Although this capability has been intensively studied almost for a half a century, magnetoreceptive structures have still not been found nor has the precise mechanism of processing magnetic information been explained. Moreover, in many animals even the purpose of this extraordinary sensory capacity remains unclear. The introductory part of the work describes two possible magnetoreception mechanisms and a special attention is given to the mechanism of light-dependent magnetoreception. This thesis focuses on the ability to identify the direction of the magnetic field vector, which is a crucial prerequisite for any magnetosensitive system to work as a compass or to be useful for spatial orientation. Besides, the work stresses the need for independent replications of data on magnetoreception – a frequently discussed topic at present. As part of the work, results of three attached articles are presented. The aim of study I was to verify previous findings and expand our understanding of magnetic alignment, the simplest behavior based on the direction of a magnetic field. The remaining two works use different diagnostic approaches to investigate and prove a link between directional magnetoreception and the mechanism of radical pairs. Study II shows that after Cryptochrome II silencing by means of RNAi, cockroaches become insensitive to changes in the direction of a magnetic field. Study III gives evidence of disruption of directional orientation in the marine crustaceans Gondogeneia antarctica along the sea-land axis after the application of a weak radiofrequency field – a phenomenon accepted as a diagnostic tool for magnetoreception based on the radical pair mechanism.
