Influence of lysogeny on the sensitivity of Staphylococcus aureus strains to polyvalent bacteriophages

Abstract:

Staphylococcus aureus je oportunní patogen, který u lidí s oslabenou imunitou vyvolává závažná onemocnění, jež mohou mít fatální následky. V současné době je léčba těchto infekcí značně problematická a to díky zvyšujícímu se počtu kmenů rezistentních k antibiotikům. Východiskem by mohla být fágová terapie, která se již řadu let aplikuje v některých východních státech, například v Gruzii, Rusku nebo Polsku. Jedním z problémů fágové terapie je rezistence vůči polyvalentním fágům, které se k této terapii používají. Tato rezistence k fágům může mít řadu příčin. Doposud byla pozornost zaměřena spíše na restrikčně-modifikační systémy, významný vliv však mohou mít i profágy integrované v genomech kmenů rezistentních k polyvalentním fágům. Tato diplomová práce se zabývá vlivem lyzogenie na citlivost kmenů S. aureus k polyvalentním fágům. Schopnost navodit rezistenci u svých hostitelů byla prokázána u několika mírných fágů. Pomocí komparativní genomiky byl identifikován gen, který by mohl být zodpovědný za navození rezistence vůči fágům, a byla navržena PCR na detekci tohoto genu.

Abstract:

Staphylococcus aureus is an opportunistic pathogen that in people with weakened immune systems raises serious disease that can be fatal. Currently, the treatment of these infections is very problematic due to the increasing number of strains resistant to antibiotics. The starting point could be phage therapy, which has been applied for many years in some Eastern countries, such as Georgia, Russia or Poland. One of the problems of phage therapy is resistance to polyvalent phages used in this therapy. This resistance to phages can have many causes. So far, the attention has been focused more on restriction-modification systems. However, the prophages integrated in the genomes of strains resistant to polyvalent phages may be important too. This thesis examines the influence of lysogeny on the sensitivity of strains of S. aureus to polyvalent phage. The ability to induce resistance in its hosts has been demonstrated in a number of temperate phages. With comparative genomics a gene has been identified that could be responsible for inducing resistance to phages, and the PCR has been designed to detect this gen.
