Study of microRNAs associated with the development and progression of selected solid tumors using modern biophysical methods

Abstract:

Úvod MiRNA jsou klíčovými regulátory fyziologických a patofyziologických procesů v lidském těle a není proto divu, že výzkumu detekčních metod pro stanovení miRNA je věnováno tolik pozornosti. Detekční metody vynechávající krok hybridizace a značení jsou žádaným podnětem pro rychlejší a levnější identifikaci cílových miRNA. I když tyto metody nemají zatím takový potenciál v uvedení do praxe díky několikanásobně menší senzitivitě oproti metodám založeným na hybridizaci s miRNA, tato práce se pokouší, prostřednictvím nových strukturálních informací o miRNA, přinést nové poznatky využitelné k rozvoji label-free metod. Metody V práci se snažíme zjistit co největší množství informací o miRNA na základě její struktury, proto byly voleny metody elektrochemické (voltametrie: LSV, CV a SWV) a metody optické (pro studium elektronových vibrací: UV-VIS a CD spektroskopie a pro studium vibrací jader: Ramanova spektroskopie a Ramanova optická aktivita). Pro doplnění fyzikálně-chemických analýz byla použita i metodika molekulární biologie – kvantitativní polymerázová řetězová reakce v reálném čase (RT-qPCR). Výsledky Pomocí voltametrie lze vyvodit závislost mezi obsahem guaninové báze a výškou jejího píku ve voltamogramu u dané miRNA. Stejně tak můžeme využít G píku pro definici cukr fosfátové páteře voltametrickými metodami. Díky spektrálním analýzám jsme schopni rozlišit jak povahu cukr fosfátové páteře, tak od sebe odlišit jednotlivé miRNA. Závěr Biofyzikální metody na bázi elektrochemie a spektroskopie jsou výborným základem pro detailní studium struktury biomolekul. Tato práce se snaží přiblížit novým technikám umožňujícím miRNA studovat a získat o nich nové informace společně s jejich strukturálním chování. Na vývoj spolehlivé metody, která by mohla detekovat jedinou miRNA bez značení, však budeme muset ještě počkat.

Abstract:

Background MiRNAs are key regulators of physiological and pathophysiological processes in the human body, and therefore it is no wonder that much attention is paid to the research of detection methods for miRNA determination. Detection methods omitting the hybridization and labeling step are therefore a desirable stimulus for faster and cheaper identification of target miRNAs. However, these methods do not have the potential yet to be put into practice due to the several times less sensitivity than those based on hybridization with miRNAs. This work attempts to approach label-free methods through new structural knowledge about miRNAs. Methods We try to find out as much information about miRNA based on its structure, therefore electrochemical methods (voltammetry: LSV, CV and SWV) and optical methods (for the study of electron vibrations: UV-VIS and CD spectroscopy and for the study of nuclei vibrations: Raman spectroscopy and Raman optical activity were chosen). Molecular biology methodology - real-time quantitative polymerase chain reaction (RT-qPCR) was used to complement the physico-chemical analyzes. Results Voltammetry can be used to determine the relationship between the guanine base content and the height of its peak in the voltamogram for a given miRNA. Similarly, we can use the G peak to define sugar phosphate backbone by voltammetric methods. Thanks to spectral analyzes, we are able to distinguish both the nature of the phosphate backbone sugar and the individual miRNAs. Conclusion Biophysical methods based on electrochemistry and spectroscopy are an excellent basis for a detailed study of the structure of biomolecules. This work aims to introduce new techniques enabling miRNAs to study and obtain new information about them together with their structural behavior. However, we will have to wait for the development of a reliable method that could detect a single miRNA without labeling.
