Characterisation of function domains of poly(A) polymerase Trf4p in vivo

Abstract:

Bakalářská práce, kterou předkládám, je zaměřena na oblast kontroly kvality RNA v eukaryotickém jádře. Jedním z významných prvků metabolismu RNA je exozom, komplex homologů exonukleas a endonukleasy. Ve své degradační činnosti je stimulován kofaktory, mezi něž se řadí i jaderný komplex TRAMP (Trf4p/Trf5p–Air1p/Air2p–Mtr4p polyadenylační komplex). Funkcí komplexu TRAMP je specificky rozpoznat vadné nebo nadbytečné RNA a ty cílit k degradaci exozomovým komplexem. Mechanismus rozpoznání RNA ani mechanismus stimulace exozomu není dosud uspokojivě prozkoumán. Cílem práce bylo charakterizovat úlohu C– a N–terminálních oblastí nekanonické poly(A)–polymerasy Trf4p, která je katalytickou podjednotkou komplexu TRAMP. Byly vytvořeny kmeny Saccharomyces cerevisiae exprimující mutantní Trf4p a tyto byly podrobeny fenotypové analýze skladby ribozomálních RNA pomocí northern blottingu. Analýza zatím nepřinesla jasné odpovědi, avšak při přípravě mutantních kmenů vyvstala zajímavá otázka toxicity N–terminálně zkráceného proteinu. Práce tak vytvořila základ pro další výzkum funkce proteinu Trf4.

Abstract:

The bachelor thesis I present here focuses on the RNA quality control in eukaryotic nucleus. Exosome, a protein complex consisting of exonucleases and endonucleases homologues, is a key player in RNA metabolism. Exosome is stimulated by a number of cofactors including TRAMP complex (Trf4p/Trf5p–Air1p/Air2p–Mtr4p polyadenylation complex). The TRAMP complex specifically recognizes aberrant RNAs and targets them for degradation by the exosome complex. The principles of RNA recognition or TRAMP mediated exosome stimulation are not well described so far. The thesis aims to characterize the role of the C– and N–terminal regions of the noncanonical poly(A)–polymerase Trf4p, the catalytic subunit of the TRAMP complex. Saccharomyces cerevisiae strains expressing mutant Trf4p were prepared and later on submitted to northern blot analysis of ribosomal RNA pattern. The research is still in progress and at this point we cannot make any conclusions. However, our experiments brought up an interesting issue of possible toxicity of the N–terminally truncated Trf4p. Therefore this work lays the foundations of upcoming Trf4p function investigation.
