Nematodes of the genus Thelazia in grazing cattle in the Czech Republic.

Abstract:

Tato diplomová práce se zabývá detekcí hlístic rodu Thelazia u paseného skotu v České republice. Parazité jsou lokalizováni v očním aparátu savců, zejména ve spojivkovém vaku či slzných kanálcích, kde způsobují dráždění tkání, nadměrné slzení a v závažných případech i ulceraci rohovky a slepotu. Vzhledem k tomu, že vývojový cyklus těchto nematod závisí na mezihostitelských vektorech, představuje nárůst průměrných teplot riziko pro jejich šíření do nových oblastí. Hlavním cílem práce bylo ověřit hypotézu o přítomnosti těchto opomíjených hlístic v tuzemských chovech v souvislosti s měnícími se klimatickými podmínkami. Práce reaguje na dlouhodobý nedostatek aktuálních informací o tomto parazitovi u skotu a usiluje o doložení jeho přítomnosti v našich podmínkách. K dosažení stanovených cílů byl realizován sběr očních vzorků a jejich následné laboratorní vyšetření, které bylo zaměřeno na průkaz těchto hlístic a získání aktuálních údajů o rozšíření Thelazia sp. v České republice. V rámci výzkumu proběhl v podzimních měsících sběr vzorků na sedmi lokalitách ve třech krajích České republiky. Celkem bylo vyšetřeno 208 očních vzorků ze 104 kusů skotu chovaných v extenzivních podmínkách ustájení. Vzorky byly odebrány bezprostředně po porážce a následně podrobeny laboratorní pitvě. Pro identifikaci druhu hlístice byla využita morfometrická analýza. Výzkumem byla potvrzena přítomnost očních hlístic rodu Thelazia u skotu v České republice. Parazité byli detekováni u dvou hostitelů a celková prevalence činila 2,08 %. Hlístice byly nalezeny v lokalitách Úročnice a Velké Popovice. Průměrná intenzita u pozitivních případů činila čtyři hlístice na jednoho hostitele. Při detailním morfometrickém vyhodnocení vybrané hlístice se potvrdilo, že nalezený jedinec se nacházel v pokročilém larválním stádiu, podle naměřených parametrů se jednalo o druh Thelazia rhodesi. Zásadním přínosem práce je první vědecky doložený nález těchto hlístic u skotu na našem území, což výzmaně doplňuje dosavadní poznatky o epizootologické situaci v tuzemských chovech. Výsledky potrzují, že thelazióza představuje aktuální riziko a zdůrazňují potřebu dalšího sledování tohoto parazita v souvislosti klimatických změn.

Abstract:

This thesis focuses on the detection of nematodes of the genus Thelazia in grazing cattle in the Czech Republic. These parasites are found in the ocular apparatus of mammals, particularly in the conjunctival sac or lacrimal ducts, where they cause tissue irritation, excessive tearing, and, in severe cases, corneal ulceration and blindness. Given that the life cycle of these nematodes depends on intermediate hosts, rising average temperatures pose a risk for their spread to new areas. The main objective of this study was to verify the hypothesis regarding the presence of these neglected nematodes in domestic herds in connection with changing climatic conditions. This study addresses the long-standing lack of up-to-date information on this parasite in cattle and aims to document its presence under local conditions. To achieve these objectives, eye samples were collected and subsequently examined in the laboratory, with the focus on detecting these nematodes and obtaining current data on the distribution of Thelazia sp. in the Czech Republic. As part of the research, samples were collected in the autumn months at seven locations in three regions of the Czech Republic. A total of 208 eye samples from 104 head of cattle raised under extensive housing conditions were examined. The samples were collected immediately after slaughter and subsequently subjected to laboratory necropsy. Morphometric analysis was used to identify the nematode species. The study confirmed the presence of eye nematodes of the genus Thelazia in cattle in the Czech Republic. The parasites were detected in two hosts, with a prevalence of 2.08% at the sites of Úročnice and Velké Popovice. The average intensity in positive cases was four nematodes per host. A detailed morphometric evaluation of a selected nematode confirmed that the specimen was in an advanced larval stage; based on the measured parameters, it was identified as the species Thelazia rhodesi. A key contribution of this study is the first scientifically documented detection of these nematodes in cattle in our country, which significantly supplements existing knowledge about the epizootiological situation in domestic herds. The results confirm that thelaziosis poses a current risk and underscore the need for further monitoring of this parasite in the context of climate change.
