Identification of pathogenic bacteria by MALDI-TOF - Matrix Assisted Laser Desorption/Ionization - Time of Flight

Abstract:

Cílem této práce je identifikace bakteriálních kmenů metodou MALDI-TOF (hmotnostní spektrometrie s laserovou desorpcí a ionizací za účasti matrice s průletovým analyzátorem), který se stal velmi rozšířeným a užitečným analytickým přístrojem v diagnostických laboratořích. Identifikace mikroorganismů metodou MALDI-TOF je charakteristická svou vysokou přesností, citlivostí a rychlostí identifikace ve srovnání s tradičními metodami. Součástí práce je také srovnání identifikace bakterií pomocí MALDI-TOF s jejich biochemickou identifikací, uskutečněná produkty řady MIKROLATest, vyhodnocení jednotlivých testů a vzájemné porovnání výsledků. Výsledky jednotlivých analýz se lišily svou přesností a jistotou identifikace. Na základě zjištěných údajů lze konstatovat, že identifikace mikroorganismů pomocí MALDI-TOF v diagnostických laboratořích velmi urychluje a usnadňuje práci. Ale v některých výjimečných případech není schopen zcela rozlišit druh daného rodu, kdy je poté zapotřebí použít doplňkové biochemické testy.

Abstract:

The aim of this thesis is the identification of bacterial strains using the MALDI-TOF method (Matrix Assisted Laser Desorption Ionization Time-of-Flight), which has become very commonly used, and is a useful analytical device in diagnostic laboratories. The MALDI-TOF microorganism identification is characterized by its accuracy, sensitivity and the speed of the identification when comapared to traditional methods. Another part of this thesis deals with comparing the MALDI-TOF bacterial identification to biochemical identification, made by the products from the MIKROLAtest line. It also deals with the evaluation of individual tests and compares the results. The results of individual analysis differed in accuracy and in the certainty of identification. Based on the data it can be stated that the identification of microorganisms using MALDI-TOF diagnostic laboratories greatly accelerates and facilitates the work. However, in some rare cases it is not able to perfectly distinguish the species of a given genus. In these cases i tis necessary to use complementary biochemical tests.
