Preparation of antibodies against defence proteins of tomato and tobaco

Abstract:

PR proteiny tvoří základní složku inducibilních obranných mechanismů rostliny. Jedná se o nízkomolekulární proteiny, stabilní při nízkém pH, termostabilní a vysoce rezistentní vůči působení proteas. V buňce se nachází ve vakuolách a apoplastu, kdy základními signálními molekulami podílejícími se na aktivaci jejich exprese jsou kyselina salicylová, kyselina jasmonová a ethylen. V rámci své biologické aktivity disponují antimikrobiální aktivitou, kontaktní toxicitou a některé z nich se pravděpodobně účastní signálních mechanismů během obranné reakce rostliny. Mezi PR proteiny patří i glukanasy a thaumatin-like proteiny patřící do PR-2 a PR-5 třídy. Obě tyto třídy proteinů vykazují antifungální aktivitu, i když se jejich mechanismy působení velmi liší. Zatímco glukanasy se podílí na degradaci buněčných stěn hyf a zároveň uvolňují oligosacharidové elicitory, které je hostitelská rostlina schopna detekovat, thaumatin-like proteiny nevykazují enzymatickou aktivitu a jejich antifungální vlastnosti jim propůjčuje jejich amfifilní charakter. Thaumatin-like proteiny jsou schopny vmezeřit se do fosfolipidové dvojvrstvy hub a permeabilizovat tak jejich plazmatickou membránu. V rámci diplomové práce byla provedena exprese rekombinantních obranných proteinů glukanasy a thaumatin-like proteinu v expresním systému E. coli ve vektoru pET32 za využití autoindukčního média. Následně byly rekombinantní proteiny purifikovány pomocí metody IMAC-FPLC. U úspěšně purifikovaných rekombinantních proteinů byla ověřena biologická aktivita a pomocí metody TSA byly nalezeny vhodné podmínky pro budoucí skladování proteinů.

Abstract:

PR proteins form basic component of inducible defence mechanisms of plants. They are low-molecular proteins, stable in low pH, thermostable and highly resistent toward effect of proteas. They are localized in cells vacuols and apoplast. Primary signal molecules participating on activation of their expression are salicylic acid, jasmonic acid and ethylen. PR proteins possess antimicrobial activities, contact toxicity and some of them are perhaps involved in defense signaling during defense mechanisms of plants. Glucanases and thaumatin-like proteins also belong to PR proteins, more specifically to families of PR-2 and PR-5 proteins respectively. Both PR-2 and PR-5 proteins exhibits antifungal activity even if their mechanism of action is quite different. While glucanases partake in degradation of hyphae cell walls and at the same time release oligosacharid elicitors, which can be detect by host plant, thaumatin-like proteins don´t exhibit enzymatic activity and their antifungal activity is due to their amphiphilic character. Thaumatin-like proteins are able to insert to phospholipid bilayer of fungi and permeabilized their plasma membrane. In framework of thesis was carrie out expression of recombinant defense proteins, glucanase and thaumatin-like protein, in express system of E. coli in vector pET32 with use of autoinduction medium. Consequently recombinant proteins were purified with help of method IMAC-FPLC. Successfully purified recombinant proteins were test for biological activity and via method TSA were found adequate conditions for future storing of proteins.
