Study of stability of antidiabetic Ezetimibe in acid and base medium

Abstract:

Tato bakalářská práce se zabývá identifikací produktu vznikajícího z Ezetimibu v alkalickém prostředí, o jehož struktuře byly doposud publikovány rozporné informace. Na základě rentgenostrukturní analýzy, 1H, 13C NMR spektroskopie a HPLC s použitím chirální kolony byl jako produkt rozkladu ve vodném NaOH jednoznačně potvrzen (2R,3R,6S)-N,6-bis(4-fluorfenyl)-2-(4-hydroxyfenyl)-3,4,5,6-tetrahydro-2H-pyran-3-karboxamid namísto chybně navržené soli 5-(4-fluorfenyl)-2-[(4-fluorfenylamino)-(4-hydroxyfenyl)methyl]pent-4-enové kyseliny. Druhá část práce se zabývá kinetikou a mechanismem bazicky katalyzovaného přesmyku Ezetimibu ve vodných roztocích terciárních aminových pufrů a v hydroxidu sodném. Na základě kinetických měření byl navržen mechanismus zahrnující postupnou tvorbu monoaniontu (alkoholátu) resp. dianiontu (alkoholátu a fenolátu) v rychlé předřazené rovnováze, které podléhají intramolekulárnímu přesmyku za vzniku zmíněného produktu. V silně bazickém prostředí (roztoky NaOH s koncentrací vyšší než 0,05 M) se začíná tvořit nový rozkladný produkt - pravděpodobně sůl 5-(4-fluorfenyl)-2-[(4-fluorfenylamino)-(4-hydroxyfenyl)-methyl]pentanové kyseliny.

Abstract:

This bachelor work deals with the correct structure elucidation of alkaline degradant of Ezetimibe which was recently misinterpreted in the literature. On the basis of X-ray, 1H, 13C NMR spectroscopy and chiral HPLC I unambiguously proved that the correct structure formed from Ezetimibe in aqueous NaOH is (2R,3R,6S)-N,6-bis(4-fluorophenyl)-2-(4-hydroxyphenyl)-3,4,5,6-tetrahydro-2H-pyran-3-carboxamide instead of previously suggested 5-(4-fluorophenyl)-2- [(4-fluorophenylamino)-(4-hydroxyphenyl)methyl]-pent-4-enoic acid. The second part of this work deals with kinetics and mechanism of base-catalyzed rearrangement of Ezetimibe in aqueous solutions of tertiary amine buffers and in sodium hydroxide. On the basis of kinetic measurements the following reaction mechanism was suggested: Mono anionic species (alkoxide) and dianionic species (alkoxide and phenoxide) are gradually formed in fast pre-equilibria which then undergo intramolecular rearrangement to give product mentioned above. Under strong base conditions (solutions of NaOH with concentration higher than 0.05 M) completely new degradation product begins to form which is the most probably salt of 5-(4-fluorophenyl)-2-[(4-fluorophenylamino)-(4-hydroxyphenyl)-methyl]-pentanoic acid.
