Application of capillary electrophoresis for beta-secretase activity assay

Abstract:

Tato bakalářská práce je zaměřena na využití kapilární elektroforézy pro studium kinetických parametrů enzymu beta-sekretasy. Jedná se o proteolytický enzym, jehož zvýšená hladina v mozkových buňkách napomáhá rozvoji Alzheimerovy choroby. Tento jev je popsán tzv. teorií amyloidových plaků, která v současné době patří mezi nejuznávanější, ačkoliv přesná příčina vzniku choroby nebyla dosud bezpečně objasněna. Inhibice beta-sekretasy by podle této teorie mohla vést k potlačení vzniku nemoci, a proto se v posledních letech usiluje o nalezení vhodného inhibitoru. Mezi nejpoužívanější separační techniky pro studium aktivity enzymů patří kapilární elektroforéza především díky vysoce účinným separacím a minimální spotřebě vzorku a ostatních chemikálií. V této práci byla kapilární elektroforéza spojena s detekcí pomocí hmotnostní spektrometrie. Ačkoliv toto spojení není zcela běžné pro studium aktivity beta-sekretasy, neboť obvykle se používá vysokoúčinná kapalinová chromatografie s fluorescenční detekcí, podařilo se nám optimalizovat vhodnou metodu pro kinetické studie tohoto enzymu.

Abstract:

This bachelor’s thesis focuses on the application of capillary electrophoresis technique for the kinetic studies of the beta-secretase enzyme. An increased level of this proteolytic enzyme in brain cells contributes to the development of Alzheimer's disease. This phenomenon is described by the so-called amyloid plaques theory which nowadays counts among the most respected theories, although the exact cause of the development of the disease has not been securely clarified yet. However, the inhibition of beta-secretase could lead to a suppression of the development of the disease and therefore many researchers strive for discovery of a suitable inhibitor in the last years. Capillary electrophoresis belongs among the most widely used separation techniques used for the activity studies of enzymes, particularly due to the high efficiency of the separations and the minimum consumption of sample and other chemicals. In this work the capillary electrophoresis was coupled with mass spectrometry detection. Even though this connection is not common for the study of the activity of beta-secretase the fluorescence detection is often used instead in combination with high performance liquid chromatography, we managed to develop a promising method for kinetic studies of this enzyme.
