Polysaccharide-based capsules prepared using FDM 3D printing technique

Abstract:

Tato práce se v teoretické části zabývá novými poznatky v oblasti přípravy tobolek z biopolymerních materiálů, s důrazem na alternativní techniky, jež otevírají perspektivní možnosti pro farmaceutický průmysl. Konkrétně se zaměřuje na kombinaci technologií hot melt extruze (HME) a FDM 3D tisku. V experimentální části byly tyto inovativní metody aplikovány na přípravu enterosolventních polysacharidových tobolek. Pro výrobu těchto tobolek byla použita kombinace polosyntetického a biopolymerního materiálu (AffinisolTM s citrusovým pektinem nebo alginátem sodným). Studován byl také přídavek plastifikátoru polyethylenglykolu (PEG) scílem snížit teplotu extruze a zlepšit vlastnosti extrudovaného vlákna s ohledem na 3D tisk. Proces přípravy zahrnoval extruzi filamentů metodou HME, následovanou samotným 3D tiskem, plněním a testováním tobolek (rozměrová analýza, disoluční testování). Tato práce přináší nové poznatky v oblasti výzkumu enterických tobolek na bázi polysacharidů pro cílený transport léčiva do oblasti kolonu. V rámci diskuze byly zhodnoceny možnosti použití připravených tobolek, jak z hlediska směrování léčiv do oblasti kolonu, tak i jako potenciálního přístupu pro fekální mikrobiální transplantaci.

Abstract:

The theoretical part of the thesis deals with novel insights into preparing capsules from biopolymer materials, emphasizing alternative techniques that open up promising possibilities for the pharmaceutical industry. Specifically, it focuses on combining hot melt extrusion (HME) and FDM 3D printing technologies. In the experimental part, these innovative methods were applied to prepare enteric-coated capsules. A combination of semi-synthetic and biopolymer materials (AffinisolTM with citrus pectin or sodium alginate) was used to produce these capsules. The addition of plasticizer polyethylene glycol (PEG) was also studied to reduce the extrusion temperature and improve the properties of the extruded filament concerning 3D printing. The preparation process included filament extrusion using the HME method, followed by 3D printing, filling, and testing of the capsules (dimensional analysis, dissolution testing). The thesis provides new insights into the research of polysaccharide-based enteric-coated capsules for targeted drug transport to the colon area. As part of the discussion, the possibilities of using the prepared capsules were evaluated in terms of targeted drug delivery to the colon area and as a possible approach for fecal microbial transplantation.
