The influence of temperature on chicken immune cells

Abstract:

Cílem práce bylo popsat vliv zvýšených teplot (41 °C a 45 °C) na buňky imunitního systému kura domácího izolované z krve, sleziny, brzlíku a Fabriciovy burzy a porovnat jejich citlivost vůči zvýšeným teplotám. Dále bylo cílem zjistit, jak tyto buňky ovlivní přídavek baicalinu a baicaleinu při různých teplotách. Buňky byly vyizolovány a následně vloženy do inkubátorů s 37 °C (optimální teplota), 41 °C (mírný tepelný stres) a 45 °C (výrazný tepelný stres). Po 3 a po 8 hodinách bylo vyhodnoceno zastoupení apoptotických a nekrotických buněk. Dále bylo hodnoceno zastoupení buněk pozitivních na CellROX a MitoSOX značící probíhající oxidativní stres. Při 45 °C došlo k významnému zvýšení buněčné smrti zejména po 8 hodinách inkubace a také zvýšení hladin oxidativního stresu. Více citlivé ke zvýšeným teplotám byly buňky sleziny a Fabriciovy burzy. Nejvyšší hodnoty viability naopak při všech měřeních dosahovaly T lymfocyty brzlíku. Po stimulaci 45 °C došlo po 8 hodinách k významným poklesům krevních CD8 lymfocytů; CD3, CD4 T lymfocytů a BU1 B lymfocytů ze sleziny; CD3, CD4 a CD8 lymfocytů brzlíku BU1 a MHC II B lymfocytů Fabriciovy burzy. Baicalin dále při 45 °C významně pozitivně ovlivnil některé parametry oxidativního stresu u buněk sleziny a Fabriciovy burzy. Baicalein naopak u buněk krve a sleziny působil spíše prooxidačně a zhoršil oxidativní stres způsobený 45 °C. Výsledkem této práce je zjištění, že populace sleziny a Fabriciovy burzy jsou více citlivé ke zvýšené teplotě, a naopak nejodolnější jsou T lymfocyty brzlíku. Baicalin a baicalein ve velmi málo případech průkazně ovlivnili projevy buněčné smrti a oxidativního stresu u jednotlivých populací buněk, a proto nelze zhodnotit jejich jednoznačně pozitivní nebo negativní vliv při zvýšených teplotách.

Abstract:

The aim of the work was to describe the effect of elevated temperatures (41 °C and 45 °C) on immune cells of the broiler chicken isolated from blood, spleen, thymus and bursa of Fabricius and compare their sensitivity to elevated temperatures. Furthermore, the aim was to find out how these cells will affect the addition of baicalin and baicalein at different temperatures. Cells were isolated and then placed in incubators at 37 °C (optimal temperature), 41 °C (mild heat stress) and 45 °C (severe heat stress). After 3 and after 8 hours, the percentage of apoptotic, necrotic cells was evaluated. Furthermore, the representation of CellROX and MitoSOX positive cells indicating ongoing oxidative stress was evaluated. At 45 °C, there was a significant increase in apoptotic cells after 3 and especially after 8 hours of incubation. The cells of the spleen and bursa of Fabricius were more sensitive to elevated temperatures. On the other hand, the highest values of viability were achieved in T lymphocytes of the thymus. There were no significant decreases in individual lymphocyte phenotypes at 41 °C. At 45 °C, there were significant decreases in blood CD8 lymphocytes after 8 hours; CD3, CD4, and BU1 B lymphocytes from the spleen; CD3, CD4 and CD8 thymic lymphocytes; BU1 and MHC II B lymphocytes of the bursa of Fabricius. Furthermore, at 45 °C, baicalin significantly positively influenced some parameters of oxidative stress in cells of the spleen and bursa of Fabricius. Baicalein, on the other hand, had a more pro-oxidative effect on blood and spleen cells and worsened the oxidative stress caused by 45 °C in some cases. The result of this work is the finding that the population of the spleen and the bursa of Fabricius are more sensitive to elevated temperature, and on the contrary, the T lymphocytes of the thymus are the most resistant. In very few cases, baicalin and baicalein significantly influenced the manifestations of cell death and oxidative stress in individual cell populations, and therefore it is not possible to ascertain their clearly positive or negative effect at elevated temperatures.
