Immunoaffinity interaction studied using electrochemical methods with regard to charged functional components

Abstract:

Tato diplomová práce se zabývá vývojem imunosenzoru s elektrochemickou detekcí. Modelový koncept tohoto biosenzoru zahrnuje interakci mezi lidským sérovým albuminem (HSA) a jeho příslušnou protilátkou. Převodník sestává ze dvou identických zlatých elektrod – nevyžadují žádnou externí referentní elektrodu, což v budoucnu umožní miniaturizaci zařízení. HSA, který představuje antigen, byl imobilizován na elektrody prostřednictvím poly-L-lysinu a citrátu za použití aktivačního činidla EDC. Imobilizovaný protein interagoval s protilátkou ve vzorku. Změny na povrchu elektrody byly zachyceny měřením elektrochemické impedanční spektroskopie (EIS) v prostředí mediátoru ferro/ferrikyanidu ([Fe(CN)6]4−/[Fe(CN)6]3−). Testování biosenzoru podalo neočekávané výsledky – elektrochemická impedance se s narůstající koncentrací přidané protilátky snížila. Diplomová práce tedy rozvíjí především možný vliv náboje jednotlivých biomolekul, který je často ignorován.

Abstract:

This diploma thesis focuses in the first place on a development of immunosensor with electrochemical detection. Its model concept covers the interaction between human serum albumin (HSA) and the corresponding antibody. Electrochemical transducer consists of two identical gold electrodes – no external reference electrode is needed. So, the measuring system could be miniaturized in future. HSA as a representative of an antigen was immobilized on the electrodes via poly-L-lysine, citrate and EDC crosslinking agent. The immobilized protein interacted with anti-HSA antibody in a sample. Changes on the surface of the electrode were followed by measurements of electrochemical impedance spectroscopy (EIS) in the environment of redox mediator ferro/ferricyanide ([Fe(CN)6]4−/[Fe(CN)6]3−). Testing of the biosensor yielded unexpected results – the electrochemical impedance decreased with increasing concentration of antibody present in a sample. The diploma thesis thus develops possible and often ignored influence of a charge of individual biomolecules involved.
