Dental genetic conditioned pathologies in historical population from Znojmo Hradiště

Abstract:

Tato diplomová práce pojednává o problematice geneticky podmíněných vad chrupu u vybraných jedinců z pohřebiště Znojmo Hradiště. Pro studium této práce byla vybrána zubní ageneze, která patří mezi nejčastější vývojové vady a postihuje okolo 22,5 % současné české populace. Pojmem zubní ageneze rozumíme nevyvinutí jednoho nebo několika zubů. Nejčastěji chybějícími zuby bývají třetí moláry a za nimi následují druhé premoláry a laterální řezáky. Odontogeneze je proces vývoje zubů, kdy docházi k interakci mezi orálním ektodermem a mezenchymem. Na vývoji zubů se podílí také velký počet signálních molekul, které mezi sebou interagují. Zubní ageneze se může vyskytovat ve sporadické formě, která bývá spojována s mutacemi v genech PAX9, MSX1 a AXIN2, anebo jako součást jiných genetických syndromů. Na základě makroskopického prozkoumání chrupu byli v této práci vybráni 3 jedinci z pohřebiště Znojmo Hradiště s potenciální zubní agenezí. U těchto jedinců byla provedena antropologická, rentgenologická a genetická analýza, která měla zubní agenezi potvrdit, nebo vyloučit. Součásti genetické analýzy mělo být dohledání konkrétních mutací, které by mohly být příčinou absence jednotlivých zubů. Pro molekulární analýzu byl vybrán gen PAX9, konkrétně exon 3, u kterého se vyskytuje nejvíce polymorfizmů spojených se zubní agenezí a který je dosud nejlépe prostudovanou části genu PAX9.

Abstract:

This thesis deals with the issue of genetically induced dentition defects of selected individuals from a burial site Znojmo Hradiště. The condition selected for this study is dental agenesis, which is the most common developmental abnormality and it affects about 22,5 % of the current Czech population. The term dental agenesis stands for not developing one or several teeth, in most cases the missing teeth are the third molars, then the second premolars and the lateral incisors. Odontogenesis is the process of the tooth development, in which there is an interaction between oral ectoderm and mesenchyme. Several numbers of interacting signalling molecules participate in the tooth development. Dental agenesis may occur in a sporadic form which is associated with mutations in PAX9, MSX1 and AXIN2 genes or as a part of another genetic syndromes. Three individuals were selected after thorough macroscopic exploration of the dentition of skeletal remains from the burial site Znojmo Hradiště, all these individuals were potentially suffering from dental agenesis. Subsequently, anthropological, radiological and genetic analyses, which should have confirmed or excluded the dental agenesis, were made. Specific mutations responsible for the lacking tooth were supposed to be traced by genetic analysis. Exon 3 was chosen for the molecular analysis of the PAX9 gene because the most polymorphisms associated with the dental agenesis occur in this exon. It is also the most studied part of the PAX9 gene.
