The roles of selected MAPK proteins in alfalfa Medicago sativa

Abstract:

Tato bakalářská práce byla zaměřena na studium vybraných mitogen-aktivovaných protein kinas. Nejdříve byla provedena in silico analýza MMK2 a MMK3 proteinů v Medicago sativa a jejích ortologů v Arabidopsis thaliana, jenž primárně slouží pro predikci vlastností těchto doposud málo studovaných MAP kinas. Následně byla realizována genotypová analýza stabilně transformovaných rostlin pro ověření přítomnosti transgenu kódujícího fúzní proteiny MMK3mRFP, mRFPMMK3 nebo MMK3GFP. V případě rostlin se značeným MMK3 proteinem pomocí GFP byla provedena také fluorescenční mikroskopie. Součástí této analýzy byla i sekvenční analýza potencionálně transgenních rostlin. Poté byla provedena transientní transformace rostlin Nicotiana benthamiana pomocí Agrobacterium tumefaciens. U transformovaných listů byla realizována mikroskopická analýza a také byl sledován vliv nadexprese fúzních proteinů na fenotyp listů. Na závěr byl analyzován vliv SIMK proteinu na fertilizaci se zaměřením na fenotyp pylových zrn. V této části bakalářské práce byla provedena statistická analýza studovaných parametrů u třech linií: linie divokého typu RSY, mutantní linie SIMKKi a nadexpresní linie GFP-SIMK, které byly následně porovnávány. Součástí studovaných parametrů pylových zrn byla vitalita, morfologie a klíčení.

Abstract:

This bachelor thesis was focused on the study of selected mitogen-activated protein kinases. First, in silico analysis of MMK2 and MMK3 proteins in Medicago sativa and their orthologues in Arabidopsis thaliana was performed, which is primarily used to predict the properties of these so far little studied MAP kinases. Subsequently, genotypic analysis of stably transformed plants was performed to verify the presence of a transgene encoding a MMK3mRFP, mRFPMMK3, or MMK3GFP fusion proteins while MMK3-GFP lines were also tested by presence of GFP. Sequence analysis of potentially transgenic plants was also part of this analysis. Afterwards, transient transformation of Nicotiana benthamiana plants was performed using Agrobacterium tumefaciens. Microscopic analysis was performed on the transformed leaves and the effect of overexpression of the fusion proteins on the leaf phenotype was also monitored. Finally, the effect of SIMK protein on fertilization was analyzed with a focus on the pollen grain phenotype. In this part of the bachelor thesis, statistical analysis of the studied parameters was performed for three lines: the wild type RSY, the mutant line SIMKKi and the overexpression line GFP-SIMK which were then compared. Parametres such as vitality, morphology and germination of pollen grains were studied.
