Validation of the method for the determination of acetone in serum/blood by gas chromatography

Abstract:

Tato bakalářská práce se zabývá problematikou stanovení acetonu v séru/krvi metodou plynové chromatografie. Aceton patří do skupiny ketolátek, jejichž stanovení hraje v soudním lékařství významnou roli. Stanovení těchto látek může být prospěšné zejména u posmrtných vzorků, přičemž analýza těchto biologických materiálů může být nápomocná pro objasnění nejasných příčin smrti. Teoretická část této bakalářské práce zahrnuje tři kapitoly, které shrnují poznatky o acetonu, plynové chromatografii a validaci metody. První kapitola se zabývá chemickými vlastnostmi acetonu, metabolismem a poruchami metabolismu acetonu, které popisují hlavně diabetes mellitus a stav hladovění. V kapitole plynová chromatografie je popsán hlavní princip metody a instrumentace plynového chromatografu. Třetí kapitola zahrnuje význam validace, popis analytických parametrů a porovnání rozdílů mezi validací a verifikací. V praktické části se nachází metodika a výsledky výzkumu. Popsány jsou zde laboratorní postupy a výsledky analytických parametrů důležité pro validaci metody. Praktickou částí bylo potvrzeno, že metoda je pro stanovení acetonu vhodná.

Abstract:

This bachelor thesis focuses on the determination of acetone in serum/blood by gas chromatography. Acetone belongs to the group of ketone bodies, the determination of which plays an important role in forensic medicine. The determination of these substances can be particularly useful in post-mortem samples, and analysis of these biological materials may be helpful in clarifying unclear causes of death. The theoretical part includes three chapters, which summarize the knowledge about acetone, gas chromatography and method validation. The first chapter deals with the chemical properties of acetone, metabolism and disorders of acetone metabolism, mainly describing diabetes mellitus and starvation state. The gas chromatography chapter describes the main principles of the method and the instrumentation of the gas chromatograph. The third chapter includes the importance of validation, a description of the analytical parameters and a comparison of the differences between validation and verification. The practical part contains the methodology and results of the research. In this part are described laboratory procedures and results of analytical parameters important for method validation. The practical part confirms that the method is suitable for the determination of acetone.
