Molecular diversity of a set of genes encoding secreted proteins involved in the plant-Meloidogyne incognita interaction

Abstract:

Rodina map-1 genů kóduje proteiny, pro které jsou charakteristické dvě vysoce konzervované repetitivní sekvence (58 aa a 13 aa). Byla popsána u hlístic Meloidogyne incognita, M. arenaria a M. javanica, rostlinných parazitů vyznačujících se partenogenetickým způsobem rozmnožování. Ačkoli funkce těchto proteinů je dosud neznámá, předpokládá se, že se účastní interakce mezi rostlinou a hlísticí. Jejich rozdílná exprese v isogenetických liniích lišících se pouze v (a)virulencí vůči Mi-1 genu naznačuje, že rozdíly v počtu repetic a jejich uspořádání mohou být právě za (a)virulenci zodpovědné. První část této diplomové práce monitoruje distribuci map-1 genů mezi v rámci kmene Nematoda. Použití PCR s dvěma páry primerů specifických pro map-1 geny odhalila přítomnost těchto genů jen u několika tropických druhů rodu Meloidogyne – kromě výše zmíněných k amplifikaci došlo u vzorků DNA z M. hispanica, M. floridensis, M. paranaensis, M. ethiopica a M. izalcoensis. Homologie byla potvrzena klonováním a sekvenováním 760 bp-fragmentů odpovídajícím map-1.1 genu, který byl přítomen u všech zkoumaných druhů. Během klonování a sekvenování byly také popsány tři nové formy map-1 genů. V rámci druhé části byla sekvenována centrální část map-1.1 obsahující repetitivní a přilehlé sekvence u více než 170 vzorků především z různých populací M. incognita, ale i z dalších druhů. Sekvenování bylo provedeno přímo z produktu PCR. Porovnání sekvencí ukázalo extrémní konzervaci této oblasti – sekvence se v rámci populace i druhu 100% shodovaly. Mezi jednotlivými druhy se na 621 bp lišily v 0 až 5 jednonukleotidových polymorfismech. Vysoká konzervace byla pozorována i u ostatních forem map-1 genu. Tato konzervace naznačuje extrémně silný selekční tlak působící na centrální oblast tohoto genu.

Abstract:

The map-1 gene family is a small group of genes encoding proteins harboring conserved 58 aa and 13 aa-repeats. It was described in M. incognita, M. arenaria and M. javanica, asexual plant parasitic nematodes. Although the function of those proteins remains still unknown, they are supposed to be involved in the plant-nematode interaction. In addition, differential expression of those genes between M. incognita near-isogenic lines differing only in the (a)virulence against the Mi-1 resistance gene suggested that variation in the number and arrangement of repeats could correlate with their ability to overcome the plant resistance. The first part of this thesis monitors the distribution of representatives of the map-1 gene family among 52 species belonging to the phylum Nematoda. PCR using two pairs of specific primers revealed these genes only in a small group of tropical root-knot nematodes species – except the previously mentioned species, amplification was observed also in samples from M. hispanica, M. floridensis, M. paranaensis, M. ethiopica and M. izalcoensis. The homology with map-1 genes was confirmed by cloning and sequencing a fragment corresponding to map-1.1 which was present in all species. During the cloning step also 3 new members of map-1 gene family were described. In the second part of the work, a fragment from the central repeat region of the map-1.1 gene from more than 170 individuals mainly from diverse populations of M. incognita and also from other Meloidogyne species was sequenced. Sequencing was performed directly from PCR products. Multiple alignments showed extreme high conservation of the central repeat region and 60 bp flanking sequences of these genes. Within population, no single nucleotide polymorphic sites were observed; between species from 0 to 5 nucleotide substitutions were observed. High conservation was observed also in other members of map-1 gene family. So high conservation suggests an extremely strong purifying selection acting on the central repeat region of the gene.
