Characterization of secretome of MSC with different tissue origin

Abstract:

The therapeutic effect of mesenchymal stem cells (MSC) is often attributed to the paracrine action of molecules secreted into the environment, the so-called secretome. The secretome, also a conditioned medium (CM), contains a number of growth factors, cytokines, macromolecules and extracellular vesicles that mediate signal transmission between cells or cell communication, but has a number of other biological activities. As part of this thesis, conditioned media of MSCs from fat and bone marrow obtained from different donors, conditioned media of MSCs reactivated by LPS, IL1β and MSCs cultured in hypoxia were prepared. By applying the secretome to overactivated THP-1 macrophages, their immunomodulatory effects (effect on TNFα expression) and inter-donor variability were monitored. The proregenerative effects were tested through the migration test (scratch assay) and the activation of the TGFβ signaling pathway. Furthermore, basic proteomic analysis was performed through the analysis of cytokines contained in the MSC secretome. The results indicate a high inter-donor variability and in the immunomodulatory action does not show a difference between tissue yield of MSCs. Preliminary results from pro-regenerative tests indicate activity in this area as well as the effect of hypoxia and MSC preactivation on the composition of the secretome.

Abstract:

Terapeutický účinek mesenchymálních kmenových buněk (MSC) je častokrát připisován parakrinnímu působení molekul sekretovaných do okolí tzv. sekretomu. Sekretom, také kondiciované médium (CM), obsahuje množství růstových faktorů, cytokinů, makromolekul a extracelulárních vezikul, které zprostředkovávají přenos signálu mezi buňkami či buněčnou komunikaci, ale má řadu jiných biologických aktivit. V rámci této diplomové práce byla připravena kondiciovaná média MSC z tuku a kostní dřeně získaná od různých dárců, kondiciovaná média z MSC přeaktivovaných LPS, IL1β a MSC kultivovaných v hypoxii. Aplikací sekretomu na přeaktivované THP-1 makrofágy byly sledovány jejich imunomodulační účinky (vliv na expresi TNFα) a mezidárcovská variabilita. Proregenerativní účinky byly testovány prostřednictvím migračního testu (scratch assay) a aktivace TGFβ signální dráhy. Dále byla provedena základní proteomická analýza prostřednictvím analýzy cytokinů obsažených v sekretomu MSC. Výsledky naznačují vysokou mezidárcovskou variabilitu a v imunomodulačním působení se neprojevuje rozdíl mezi tkáňovým zdojem MSC. Preliminární výsledky z proregenerativních testů naznačují spíše aktivitu v této ob-lasti a také vliv hypoxie a preaktivace MSC na složení sekretomu.
