Raman spectrometry of the urinary stones

Abstract:

Močová litiáza je onemocnění, které má vysokou prevalenci i incidenci ve vyspělých a rozvíjejících se zemích. Litiáza může mít souvislost s metabolickou poruchou, která může být projevem nebo i důsledkem nemoci, a podrobný rozbor močového kamene může přispět k odhalení a včasnému řešení. Ramanova spektroskopie je stále málo používaná metoda pro identifikaci močových kamenů především z důvodu vysoké fluorescence a nestanoveného postupu přípravy vzorku, který by fluorescenci mohl ovlivnit. Tato diplomová práce je zaměřená na nalezení optimálního nastavení přístroje, nalezení vhodné formy vzorku pro měření na spektroskopu a analýzu fluorescence. Pro naplnění cíle posouzení kvality spektra při měření močových konkrementů byla porovnána výsledná data měření laserů o vlnových délkách 488 nm, 532 nm, 633 nm a 785 nm v identickém bodě fosfátové fáze. Resumé analýz bylo dále ověřeno měřením třech fází (fosfátová, oxalátová a organická fáze), přítomných v devíti močových konkrementech. Výsledná spektra jsou podkladem pro zhodnocení použitelnosti metody pro rutinní činnost laboratoří a vědecký výzkum. Součástí práce je aplikace metod pro snížení fluorescence a fluorescenčního šumu, jako jsou bělení peroxidem vodíku v koncentraci 3%, 96% alkoholem, UV světlem a fotobělení osvitem vzorku.

Abstract:

Urine lithiasis is a disease with high prevalency and incidency and developed and developing countries. Lithiasis might have the coherence with metabolic defect, and might be the consequence or even the expression of the disease and detailed analysis of the urinary stone can contribute to disclosure and timeous solution. Raman spectrometry is still not very common method for urines stones identification mainly because of high fluorescence and non-standard of preparing the samples for measurements which could significantly affect the results of Raman spectra. This diploma thesis is focused on finding the optimal setting of spectroscopy device, finding a suitable sample form for measurement on a spectroscope and fluorescence analysis. To fulfill the goal of spectrum quality assessment in the measurement of urinary stones, the resulting data of laser measurements at wavelengths of 488 nm, 532 nm, 633 nm and 785 nm at the identical point of the phosphate phase were compared. The summary of the analyses was further verified by measuring the three phases (phosphate, oxalate and organic phases) present in the nine urinary stones. The resulting spectra are the basis for evaluating the applicability of the method for routine laboratory activities and scientific research. Part of this diploma thesis is the application of methods to reduce fluorescence and fluorescent noise, such as bleaching with hydrogen peroxide at a concentration of 3%, 96% alcohol, UV light and photobleaching by exposure of the sample.
